Aim:1. To explore the correlation between the concentration of serum HBsAg and human cervical cancer oncogene(HCCR), alpha fetal protein(AFP).2. To explore the correlation between HCCR and alanine transaminase(ALT), glutamyltranspeptidase (GGT), alkaline phosphatase (ALP) in serum of hepatitis B virus(HBV) related liver diseases.3. To detect the expressional level of HCCR mRNA in primary hepatocellular carcinoma(PHC) and explore its clinical significance.Methods:1. To study the correlation between the concentration of serum HBsAg and HCCR, AFP, serum samples of400cases patients with HBsAg and100cases healthy people were collected. According to the concentration of HBsAg, the serum samples of400cases patients with HBsAg were assigned into four groups:group A of87cases(0.5ng/ml~150ng/ml), group B of98cases(151ng/ml~300ng/ml), group C of113cases(301ng/ml~450ng/ml), group D of102cases(>450ng/ml). Enzyme-linked immuno sorbent assay was used to detect HCCR. Chemoluminescence was used to detect HBsAg and AFP. And to analyze the correlation between the concentration of serum HBsAg and HCCR, AFP.2. Studying the correlation between serum HCCRand ALT, GGT, ALP in HBV related liver diseases:Among of the400cases patients with HBsAg, there were101cases with PHC,116cases with hepatocirrhosis and183cases with hepatitis. To detect HCCR, ALT, GGT and ALP of the400cases patients with HBsAg and100cases healthy people respectively and. Then to analyze differences and correlation between groups3.Reverse transcription polymerase chain reaction was used to detect and analyze the expressional level of HCCR mRNA in PHC tissues, surrounding non-tumor tissues, liver cirrhosis and their peripheral blood mononuclear cells,Results:1. When HCCR and AFP in the groups of different concentration of serum HBsAg were compared with those in the control group, their levels were different significantly(P<0.05). The levels of serum HCCR and AFP increased gradually with the increase of the concentration of HBsAg and the level of HCCR was positively correlated with the level of AFP (P<0.01).2.Positive rate of HCCR and AFP(consider HCCR>15mg/ml、 AFP>9ng/ml as positive) in the groups of different concentration of serum HBsAg was significantly increased with the increase of concentration of HBsAg(x2=142.40, P<0.01; x2=130.76, P<0.01).3. The levels of serum HCCR, ALT, GGT and ALP in different liver disease groups were significantly higher than those in control group(P<0.01). The levels of serum HCCR, GGT and ALP in PHC were significantly higher than those in hepatocirrhosis, hepatitis and control group(P<0.05). The levels of serum ALT in PHC, hepatocirrhosi and hepatitis were significantly higher than those in control group, but there was no statistically significant difference among the three groups (P>0.05).4.The analysis of the correlation between serum HCCR and ALT, GGT, ALP in HBV related liver diseases showed that there were no relativity between HCCR and GGT, ALT, ALP(r=0.04, r=0.02, r=0.07, P>0.05). There were positive relativity between HCCR and ALT,GGT, ALP in PHC and hepatitis group(r=0.66, r=0.55, r=0.86, P<0.05; r=0.50, r=0.37, r=0.26, P<0.05).5. The relative expression levels of HCCR mRNA in the PHC tissues were0.83±0.10, and which in surrounding non-tumor tissues and the hepatocirrhosis tissues were0.12±0.07and0.57±0.12. The difference in the three groups had significantly statistical significance(P<0.05). The relative expression levels of HCCR mRNA in the PBMC of PHC patients were0.55±0.06and which in the hepatocirrhosis patients were0.34±0.04, while in the normal ones were not detected their expression. The difference had significantly statistical significance(P<0.05). Conclusions:1. The levels of serum HCCR and AFP increased gradually with the increase of the concentration of HBsAg and the level of HCCR was positively correlated with the level of AFP, which showed the level of HCCR reflected inflammatory lesions of liver cells and the extent of cancer.2. The levels of serum HCCR, ALT, GGT and ALP in patients with different HBV related liver diseases were higher than the control group, which showed that the liver function was damaged. Detecting HCCR, ALT, GGT and ALP meanwhile can help evaluate liver cells damage, the prediction of pathogenetic condition and the differential diagnosis of liver diseases.3.The expressional levels of HCCR mRNA in patients with PHC tissues, hepatocirrhosis tissues and their PBMC were up-regulated significantly, which showed that HCCR may participated in the occurrence of PHC and was related with the evolution of malignant change in the PHC.
