| ObjectiveIn this study, by verifying miRNA-29expression in pancreaticcancer, and its role in pancreatic tumor cell migration; comparepancreatic cancer patient’s tumor tissue and normal pancreatic tissuemiRNA expression by upregulating gene expression and down-regulationof miRNA-29, to prove spectrum. Initial screening expression markedlyabnormal miRNA-29c molecules in pancreatic carcinoma and itsvalidation by RT-PCR, and speculated that the mechanism of tumorigenesis and metastasis. miRNA the unique exception expression patterns inpancreatic cancer may be tumor diagnosis, clear high-risk and low-riskpopulations, disease prognosis, providing valuable information for cancerprevention and prediction of response to treatment, to provide a theoretical basis for the early diagnosis of pancreatic cancer.Research MethodsCollected after surgical resection of pancreatic cancer tissue andadjacent normal pancreatic tissue20pairs in adjacent normal tissue selected from the cancer tissue mass in the edge of at least3cm above thepancreatic tissue adjacent pancreatic tissue. Real-time fluorescencequantitative detection were extracted RNA, reverse transcription, andmiRNA-29. Analysis of its expression and tumor size, TNM stage,histological type, lymph node metastasis, age, gender, and other clinicalpathological features. ResultThe real-time RT-PCR was used to detect the sensitivity and specificity of the expression of miR-29. miR-29expression in pancreatic cancertissues was significantly higher than the adjacent tissues (P <0.001);expression of miR-29higher TNM stage, lymph node metastasis,histological type, age, and gender-related (each of grouping between P<0.01and <0.05); expression of miR-29expression in pancreatic cancertissue and adjacent pancreatic tissue between there is a statisticallysignificant difference (P=0.029).ConclusionsmiR-29is highly expressed in pancreatic carcinoma, its level may berelated to the occurrence of pancreatic cancer, metastasis inhibition hassome correlation provides theoretical basis for the early diagnosis ofpancreatic cancer. |
