Pentenyl flavonoids are a kind of flavonoid compounds, which has unique characteristics for having isoamyl alkenyl side chain on flavonoids skeleton Research shows that different pentenyl flavonoids have blood pressure falling, antibacterial、antioxidant、anti-tumor and anti-hiv bioactivity. According to the polarity, mainly including larger water-soluble flavonoids and smaller polarity pentenyl flavonoids. Literature reported the content of pentenyl flavonoids of licorice is low but has a variety of biological activity.This topic selected the licorice as the research object, studies the water-soluble flavonoids and pentenyl flavonoids extraction and purification process and quality control, further through the pathological model, I diabetes and II diabetes mice model, to study the water-soluble flavonoids and pentenyl flavonoids.(1) using HPLC analysis method, contents of licorice water soluble licorice flavonoids parts of measurement analysis, uv-vis spectrophotometry of water-soluble flavonoids in liquorice and pentenyl flavonoids content determination of analysis. By HPLC-MS method of water-soluble flavonoids and pentenyl flavonoids part quantitative analysis of flavonoids.(2) the experiment selected the best extraction and enrichment of water-soluble flavonoids process is:the water decoction method is used to extract, and solid-liquid ratio1:8, extracting time2h, extraction times2, pumping PIPO-01high-pressure resin, collect35%ethanol elution liquid, dry enrichment; pentenyl flavonoids with95%ethanol reflux extraction of liquorice residue, solid-liquid ratio1:8, extracting time2h, extraction times2, to extract the early15BV of chloroform extract and heating reflux extraction,70℃, reflux extracting3times, each time2h, vacuum drying.(3) through the pathological model, Ⅰ diabetes model and Ⅱ diabetes model of water-soluble flavonoids in liquorice parts with different pentenyl flavonoids effect of the flavonoids were studied. Delivery time for eight weeks, the experiment shows that the water-soluble flavonoids parts of a licorice flavonoids and isoamyl alkenyl place although no hypoglycemic effect on the normal mice, but can improve the gl tolerance of normal mice; Had no hypoglycemic effect on I diabetes model, but can improve the activity of I diabetes mice; Model mice with diabetes type Ⅱ has obvious hypoglycemic effect, also have certain lipid-lowering effect, also can improve Ⅱ diabetes glucose tolerance in mice model.
