| Objective VEGF and its receptor FLK1observed in rat liver afterischemia-reperfusion liver regeneration expression and distribution,Explore theprocess of liver regeneration molecular mechanism。Methods40healthy adult SD rats,were randomly divided into two groups,35rats in model group,5rats in normal control group,with10%chloral hydrate in the0.3~0.35ml/100g were anesthetized by intraperitoneal dose,model group weretriple blocking hepatic pedicle occlusion20min underwent liver majority (about70%)resection。Open the control group were anesthetized by intraperitoneal not block thehepatic blood flow,direct access to the normal liver tissue after death。Model grouprespectively after3h,6h,24h,2d,3d,5d,7d after the rats were killed,saidremnant liver specimens cut1×1×1cm liver tissues were fixed in10%neutralformalin fixative wet weight after,routine histological processing,after theparaffin-embedded serial sections,H-E staining of histological changes in the liver,immunohistochemical PV-9001ultra-sensitive two-step method to detect normal livertissue and model group each time point FLK1VEGF and its receptor expression andchanges in liver regeneration。Results Compared with normal control group,visible in the model group were moreinflammatory cells in liver tissue,cord between the liver cells and endothelial cells in the portal area of edema,obviously enlarged liver sinusoid,hepatic sinusoidleukocytosis, mainly neutrophils。VEGF expression is weak in normal liver tissue,positive cells were mainly endothelial cells portal area,a small number,surroundingliver cells did not express。Model group after3h-6h,VEGF expression increased,VEGF-positive cells was significantly increased after1d time,mainly around theportal areaï¼›VEGF positive reaction peaked at3d,whole liver lobule cells werepositive,and the closer the stronger central venous positive reactionï¼›After5d,VEGFexpression began to decline,recovered to normal control levels at7d。VEGF receptorFLK1in normal liver tissue was negative。Model group after3h,liver cells beginFLK1weak positive reactions,the positive reaction is mainly distributed in the livercell membrane and cytoplasm,expression peaked time24h,began to decline after2d time,returned to normal levels at7d。Conclusion There is a difference in time and space that liver regeneration processexpression and distribution of VEGF and receptor FLK1,probably by stimulatingendothelial cell divisionã€proliferation and induce angiogenesis after VEGF binding toits receptor FLK,thereby promoting liver regeneration。... |
