| Objective: the miRNA is a kind of length of about22nucleotides encodingsingle small RNA that they through the principle of complementary baseand its target genes of the3’end of the mRNA molecule non-coding region(3’ UTR), mediated translation inhibition of micrornas molecular targetsor cut off the degradation. Through this way of transcriptional genesilencing, that lets its important function in regulating gene expression.At present, quite a number of studies have found that in the process ofhuman blood tumor occurrence and development of a variety of micrornasplay a respectively proto-oncogenes or the role of tumor suppressor genes,they are in the different development stage of expression in hematopoieticstem cells and blood cells can regulate the growth, development,differentiation of the blood cells.This study of miR-223to study theexpression in children with acute lymphoblastic leukemia, by studying theexpression of miR-223in ALL, analysis of miR-223and the riskclassification of children acute lymphoblastic leukemia and explore itsclinical significance.Methods: this study selected the children’s hospital of Kunming inOctober2012to December2013,44cases of hospitalized children withacute lymphoblastic leukemia, including ALL the dangerous of17cases ofchildren (SR), dangerous (IR) in children with15cases,12cases childrenwith high-risk (HR) and control group were selected20cases of ITPchildren, ALL specimens using the subjects bone marrow or peripheral blood,and extracts of total RNA, again through specific primers cDNA synthesis,using qRT-PCR analysis of specified gene miR-223test, between different groups of cycle threshold (Ct) comparing the miRNA-223relativeexpression of quantitative analysis, by using the principle of thestatistical analysis between the different groups are the miRNA-223expression whether there is a difference, and combined with clinical dataanalysis of miR-223in ALL diagnostic classification and clinicalsignificance.Results: through the qRT-PCR method, which can be observed betweenthe miRNA-223in each group are expressed, calculate the relativeexpression of groups when the miRNA-223significantly high expression inITP children, the other (0.28±0.30) in the intermediate risk category,the moderate group (0.08±0.02), in the high-risk group (0.04±0.01),with the increase of risk of ALL children, the expression of miRNA-223,after statistics analysis, the difference between groups werestatistically significant (P<0.05),suggesting the miRNA-223in ALLchildren with risk classification has certain implications.Conclusion: the relative expression of miRNA-223measured through theqRT-PCR cycle threshold (Ct) were analyzed, and adopt the method of2ï¼â–³CT’were analyzed, and theâ–³CT’=Ct purpose gene-Ct control genes,the results suggest the higher the risk of ALL, the expression of miRNA-223in the body is lower, after a group of comparison between this kind ofits difference was statistically significant, indicating ALL disease andrisk classification is related to the expression of miRNA-223lower, andsuggested the miRNA-223May exist as a kind of tumor suppressor genes,exert its biological function in the body. |
