Part I Apoptosis effects of high glucose on HUVECObjectives:observe the apoptosis effects of high glucose on HUVECMethods:HUVEC were cultured with25mmol/L glucose DMEM medium (negative control),50mmol/L glucose DMEM medium,75mmol/L glucose DMEM medium,100mmol/L glucose DMEM medium and75mmol/L mannitol+25mmol/L glucose DMEM medium (positive control) for48h, the morphological changes of cells were observed under the microscope and the cell viability (MTT) was detected. Apoptosis was measured in each group and the expression of protein of CD98hc, p38, INK was analyzed.Results:The morphology differences of HUVEC cultured in different concentrations of glucose were significant. The correlation of HUVEC apoptosis and glucose concentration was positively. The correlation of expression of CD98hc and glucose concentration was negatively. The correlation of JNK and glucose concentration was positively. There was no significant correlation between expression of p38and glucose concentration.Conclusions:The expression of protein CD98hc reduce in HUVEC apoptosis process, and there is no significant difference of expression of protein p38, the expression of JNK was significantly increased. These results suggest that high glucose may cause HUVEC apoptosis via activate JNK subtribe of MAPK pathway. Part II Protective effects of neuregulin on HUVECObjectives:Observe the protective effects of neuregulin on HUVEC.Methods:HUVEC were cultured with25mmol/L glucose DMEM medium+10ng/ml,100ng/ml and1000ng/ml neuregulin (negative control),75mmol/L glucose DMEM medium+10ng/ml,100ng/ml and1000ng/ml neuregulin and75mmol/L mannitol+25mmol/L glucose DMEM medium+10ng/ml,100ng/ml and1000ng/ml neuregulin (positive control) for48h, the morphological changes of cells were observed under the microscope. Apoptosis was measured in each group and the expression of protein of CD98hc, p38, JNK was analyzed.Results:Compared with the control group, the morphology differences of HUVEC cultured in different concentrations of neuregulin were significant. In the experimental group, the correlation of HUVEC apoptosis and neuregulin concentration was negatively. The correlation of expression of CD98hc and neuregulin concentration was positively, and the correlation of JNK and neuregulin concentration was positively. There was no significant correlation between expression of p38and neuregulin concentration.Conclusions:Neuregulin protects HUVEC may via activating CD98hc through MAPK pathway.
