| OBJECTIVE:Thought observating the effect of Schisandra Chinensis Mixture (SM) on the protein expression of WT1(Wilms’tumorl),and the mrna expression of Nephrin and Podocin in renal cortex of mice with adriamycin-induced nephropathy (ADN),to explore the possible mechanisms of SM decreases proteinuria.METHODS:Twenty-four male Balb/c mice were randomly divided into three groups:normal control group, ADN model group and SM-treatment group.The mouse model of ADN was induced by a single tail-vein injection of Adriamycin (ADR) lOmg/kg. Then to observe serum albumin(ALB)ã€body weightã€right renal weight and urine album excretion (UAE). Furthermore, the morphological changes of renal tissues were observed under microscope. The protein expression of WT1in renal cortex were determined by Western blotting.The glomerular expressions and distribution of WT1was observed by immunofluorescene method.The mRNA expression of Nephrin and Podocin in in renal cortex was determined by RT-PCR method.RESULTS:There was a significantly lower level of ALB (20.160±0.626) and in model group than that of control group and treated group (P<0.05). There was a significantly higher level of UAE(70.453±0.603) in model group than that of control group and treated group (P<0.05). There were significantly higher protein expression of WTl,and the mRNA expression of Nephrin and Podocin in normal control group and SM-treatment group than ADN model group (å‡P<0.01).The expression of WT1in glomeruli measured by immunofluorescene was normal in control group,while it was focal absence in AND mol group,and slightly absence in SM-treatment group.CONCLUSION:After treatment with SM, The protein expression of WT1, the mRNA expression of Nephrin and Podocin were up-regulated, which may safeguard podocyte,and maintain integrality in structure and function of the glomerulor base-ment membrane(GBM),and alleviate glomerulosclerosis, therefore SM reduce proteinuria of mice with AND. |
