Objectives:To observe the effects of the over-expression of HOXB4on cell proliferation and cellcycle.Methods:The recombinant over expression vector of HOXB4was leaded into human cartilageendplate stem cells. The expression of HOXB4was detected by PCR and Western Blot,CCK8and FACS analysis were used to determine the effects on cell proliferation and cellcycle respectively.Results:In human cartilage endplate stem cells transfected with the over-expression HOXB4vector, the expression of HOXB4was increased notably, and cell proliferation wasincreased significantly(P<0.05). FACS analysis showed that after the over-expression ofHOXB4, the population of S phase was decreased from29.27%to13.8%(P<0.05).Conclusions:HOXB4over expression in human cartilage endplate stem cells could increase its cellproliferation, decrease the population of its S phase. Therefore, in human cartilage endplatestem cells, HOXB4may be involved in hindering its degeneration, making it a new studyorientation of intervertebral disc degenerative diseases.
