Objective: To consummate the pharmacognosy identification database ofAconitum L.medicinal plants produced in Yunnan;Morphology,chemical compositionsanalysis and molecular technology are exploited to study on13kinds ofRanunculaceae Aconitum L. medicinal plants produced in Yunnan.To screening for thegroups and producing area of plants which have the higher bulleyaconitine A.Thisstudy is in order to lay the foundation of the protection and further domestication ofbulleyaconitine A raw material medicine germplasm resources;Preliminarilyanalysizing the distribution of ditepenoid alkaloids in Aconitum L.medicinal plantsproduced in Yunnan,which is providing the scientific basis of searching new resourcesand important significance to guide reasonable clinical medication and ensure thesafety of using clinical medication.Method: In this subject,DNA molecularsequencing technique is applied to identify the internal transcribed spacer of AconitumL.medicinal plants produced in Yunnan;Morphological characteristics analysize isused to calculate the root biomass of Aconitum L.medicinal plants produced inYunnan, and determination of bulleyaconitine A by high performance liquidchromatoagraphy (HPLC) is used to seek out the plant species of higherbulleyaconitine A.While detecting the content of the main diterpenoid alkaloids, thatprovides guidance for the development of new pharmaceutical resources.Result: Inthe research, the dendrograms of nine kinds of Subgen. Aconitum plants produced inYunnan are drawn by the analysis of rDNA-ITS sequences,we found that the testresults consistent with classical taxonomy conclusions.Screening for bulleyaconitineA raw material plant,we found that the root biomass of Aconitum hemsleyanumvar.circinatum (wild species) ranks in the top and it has the highest percentage ofbulleyaconitine A in all the test samples.The test results alse show that the ditepenoidalkaloids of A.hemsleyanum var.circinatum is the highest by detecting the content ofditepenoid alkaloids in Aconitum L.medicinal plants produced in Yunnan.Conclusion: DNA molecular technology is feasible for the identification of Aconitum L.medicinalplants produced in Yunnan.According to comprehensive analysis the results ofbulleyaconitine Aand bioavailability,we could take A.hemsleyanum var.circinatum asbulleyaconitine A raw material plant,which has certain advantages,simultaneouslyA.hemsleyanum var.circinatum may aslo becomes a new resource of analgesics andanti-inflammatory drugs.
