| ObjectiveThe emergence of malignant pleural/peritoneal effusion(MPE) is an importantfactor of poor prognosis in cancer patients.The imbalance of Th1and Th2cytokinesis one of the immunological mechanisms in the development of MPE.This study istrying to explore the expression of cytokines in MPE and their correlation withefficacy by measuring the expression levels of these cytokines before and aftertreatment with Mannatide.MethodsInject Mannatide into pleural/abdominal for the patients with MPE.Collect MPEbefore and after treatment.Then use Real-Time PCR to detect mRNA of thesecytokines in mononuclear cells(MCs).Detect the levels of cytokines in supernatant ofMPE by ELISA.Compare the differences of these cytokines between effective andnoneffective groups.ResultsIn MPE,the expressions of IL-10, TGF-and IFN are at high levels>100pg/mL.But the expressions of TNF-and IL-12are at low levels <15pg/mL Ineffective group,the expression of IL-10increases significantly after treatment both inMCs and supernatant(n=15,P<0.05).But the expression levels of IL-12,TNF-TGF-and IFN did not change significantly(n=15,P>0.05).At the same time,all of thesecytokines above didn,t change significantly after treatment in noneffectivegroup(n=10, P>0.05).ConclusionIL-10plays an important role in regulating immunity in MPE.Mannatideimproves the expression of IL-10in MPE to achieve theraputic efficacy.Thus, IL-10can serve as an immunity adjuvant in treating MPE in the future. |
