| Objective To explore the effect and its mechanism ofSarcandra glabra Nakai on radiation pneumonnopathy in miniature pigsirradiated with60Co γ-rays. Methods A total of60miniature pigs wererandomly divided into blank control group, irradiated group with15Gyof60Co γ-rays,and intervention group administered with Sarcandraglabra Nakai at the dosage of0.3g/kg before irradiation with15Gy. At4,8,12and24weeks post-irradiation, the respiratory rate and weight wereobserved, and a portion of the right lung tissue was taken out to conductHE and Masson staining examination, while the expressions of TGF-β1and TNF-α as well as the content of hydroxyproline(HP) were detected.Results The respiratory rate, the lung coefficient and the content of HP inirradiated group showed an increasing trend at4,8,12and24weekspost-irradiation, and significantly higher than two other groups (F呼=21.035, F肺=146.014, F羟=32.610, P<0.05), Those indicators wereindistinguishable between blank control group and intervention group at4and8weeks post-irradiation (F呼=0.055, F肺=2.456, F羟=5.581, P>0.05).While the indicators of the irradiation group were significantly higher than blank control group’s (F呼=91.897, F肺=93.149, F羟=83.487, P<0.05)at12weeks and24weeks post-irradiation. It was showed in thepulmonary histopathological observation that no inflammatory responseor fibrosis were found in blank control group and intervention group at4and8weeks post-irradiation. But the pulmonary fibrosis was found inintervention group at12and24weeks post-irradiation which issignificantly milder than the intervention group’s. And the levels ofTGF-β1, TNF-α in intervention group was negative at4and8weekspost-irradiation.while it was low expression at12weeks and24weekspost-irradiation which was lower than the intervention group’s.Conclusions Sarcandrae could have protective effect againstradiation-induced lung injury in miniature pigs, which might be involvedin its inhibition to the expression of TGF-βor TNF-α and HP. |
