Effect Of TFL On The Expression Of PDGF And TNF-α Of Hepatic Fibrosis In Rats

Objective: To investigate the effect of total fiavone of Litchi ChinensisSonn on the expression of PDGF and TNF-αof hepatic fibrosis in rats.Methods: dimethylnitrosamine (dimethylnitrosamine, DMN) plateletderived growth factor（PDGF）and tumor necrosis factor （TNF-α）influenceand its mechanism of action. Methods: sixty SD male rats randomly dividedinto six group, blank control group, model group, silymarin(SIL)-treatedgroup(50mg kg-1d-1), TFL high-dose group (400mg, kg-1d-1), TFLmiddle-dose group (200mg, kg-1d-1) and TFL low-dose group (100mg, kg-1d-1). The animal model of hepatic fibrosis was pprepared by DMNintraperitoneal injection of SD rats for four weeks, at the same time of produceliver fibrosis animal models, model group were gavaged with normal saline(5ml Kg-1d-1). SIL treated as gavaged with SIL (50mg Kg-1d-1). TFLhigh-dose treatment group was gavaged with TFL (400mg Kg-1d-1). TFLmiddle-dose treatment group was gavaged with TFL (200mg, kg-1d-1) TFL low-dose treatment group was gavaged with TFL (100mg Kg-1d-1). After4weeks of treatment, the rats were sacrificed and the serum and liver tissuesamples were collected.The serum level of hyaluronic acid (HA), laminin (LN),Ⅲ procollagen (PCⅢ) was determined by RIA. The liver histopathology wasobserved with light microscopy after HE and masson staining. By polymerasechain reaction (PCR) method to detect SIL group and TFL groups PDGF, TNFfactor expression level change, analyzed the data with SPSS11.0statisticallysoftware.Results: As compared with model group, the serum levels of HA，LN andPC Ⅲ were significantly decreased (P<0.01), and the liver fibrosis wassignificantly improved in the rats treated by high-dose TFL, middle-dose andSIL; the serum levels of ALT and AST were obviously decreased (P<0.05) andthe expressions of PDGF and TNF-α were decreased in the liver of the ratswhich were given high-dose TFL (P<0.05). The serum levels of HA、LN andPCⅢ have no significantly difference between the TFL middle-dose treatmentgroup and SIL group (P＞0.05). And they have no significantly difference in thestages of liver fibrosis, the expressions of PDGF and TNF-α.Conclusion: TFL can cut down the level of PDGF and TNF cytokineexpression in liver fibrosis rats. And it has obvious effect of anti-rat liverfibrosis.
