Objective:To study the relationship between CXCR5+CD4+T cells three subsets (CXCR3+CCR6Th1like, CXCR3-CCR6-Th2like. CXCR3"CCR6+Th17like) in peripheral blood and MG. To explore the pathogenesis of MG.Methods:Percentages of CXCR5+CD4+T cells three subsets in peripheral blood of experimental subjects were analyzed by Flow Cytometry(FCM). Those experimental subjects containded20MG patients who had never received any drugs(untreated MG group),16MG patients who had received steroid therapy(treated MG group) and20healthy people(normal group). Differences of three subsets’percentages of those groups were compared. Meanwhile, differences of three subsets’ percentages of5MG patients at the time of before steroid therapy and after steroid therapy were analyzed. According to thymus differences, untreated MG group was divided into thymus normal groupã€thymic hyperplasia groupã€thymoma group.And differences of three subsets’ percentages of those groups were compared. On the basis of affected muscles, untreated MG group was divided into ocular type and generalized type. And compared differences of three subsets’percentages between the two types. Besides, according to MGFA classification, untreated MG group was classified. Also compared differences of three subsets’percentages of these types. Moreover, the relationship between percentages of CXCR5+CD4+T cells three subsets of20uncured MG patients and clinical indexes were analyzed.Results:â‘ Compared with normal group, percentage of Th1like subset and Th17like subset of untreated MG group were decreased(P<0.05); But percentage of Th2like subset was increased (P<0.05).â‘¡Percentage of Th1like subset and Th17like subset of untreated MG group had no significant difference compared with treated MG group(P>0.05); However,percentage of Th2like subset was decreased (P<0.05) After steroid therapy, percentage of Thl like subset was increased (P<0.05); Percentage of Th2like subset was decreased (P<0.05); In addition percentage of Th17like subset had no significant difference(P>0.05).â‘¢Frequencies of CXCR5+CD4+T cells subsets had no significant difference among thymus normal groupã€thymic hyperplasia group and thymoma group(P>0.05).â‘£Percentage of Thl like subset and Th17like subset in generalized type had no significant differences compared with ocular type (P>0.05);However,percentage of Th2like subset was increased (P<0.05). In addition,frequencies of CXCR5+CD4+T cells subsets had no significant difference among different MGFA classifi-cation (P>0.05).⑤No significant differences were found between percentages of CXCR5+CD4+T cells subsets and age of onsetã€disease courseã€immune full set (P>0.05).Besides, percentage of Thl like subset and Th17like subset had no relationship with QMG score and the concentration of AChR-Ab in serum (P>0.05);However, percentage of Th2like subset positively correlated with QMG score and the concentration of AChR-Ab (P<0.05).Conclusion:Percentage of Th2like subset of MG patients was increased compared with normal group, and the percentage was decreased after steroid therapy; Percentage of Th2like subset of generalized type was higher than ocular type; Besides,percentage of Th2like subset positively correlated with QMG score and the concentration of AChR-Ab in serum. |
