Objective:To explore the expression of neural epithelial cells transforming gene1(Net-1) and their significance in human non-small cell lung cancer (NSCLC)Materials and Methods:Real-time quantitative PCR of tissue samples collected from the time of the Second Xiangya Hospital cardiothoracic surgery of Central South University,consists of surgical removal of tissue from NSCLC lung cancer patients from October2009to April2012. A total of64cases, Western-blotting and immunohistochemistry’s samples are come from the Third Xiangya Hospital of Central South University surgical removal of tissue from NSCLC lung cancer patients from2012.2to2013.6, A total of20cases with no preoperative radiotherapy and chemotherapy,each sample contain the lung tissue and para-carcinoma normal tissues (the tissues cancer does not invade) using2cases of normal lung tissues as control group in Immunohistochemical. The expression and its clinical significance of Net-1in non-small cell lung cancer (NSCLC) is detected by real-time quantitative PCR, Western-Blotting,Immunohistochemical.Result:The real-time quantitative PCR showed that the expression of NSCLC lung cancer tissues Net-1gene mRNA is higher than its para-carcinoma tissues.The expression of Net-1mRNA in poorly differentiated cases is higher than well differentiated cases,it has positive correlation with histological differentiation (P<0.05), the Ⅲ phase and Ⅱ phase patients was significantly increased compared with Ⅰ Phase (P<0.05),it has positive correlation with TNM classification, in NSCLC tissues,there is no relationship between the expression of Net-1mRNA and age, gender, smoking history, tumor size, pathological type, lymphatic metastasis (P>0.05).Western blotting showed the expression of Net-1protein in NSCLC lung cancer tissues was significantly higher than para-carcinoma tissues, the results of Western blotting associated with clinical data analysis showed that the expression of Net-1protein is associated with differentiated degree,TNM classification and lymphatic metastasis (P<0.05). the expression of Net-1protein has no relationship between age, gender, smoking history, histological type, tumor size (P>0.05).The results of Immunohistochemistry shows that the Net-1expression in normal lung tissues were negative, Net-1mainly express on the cytoplasm and cell membrane in NSCLC tissues, cancerous tissue expression is higher than normal lung tissues.Associated with clinical data analysis showed that the expression of Net-1protein is associated with TNM classification, differentiated degree, lymphatic metastasis (P <0.05). There is no relationship between the expression of Net-1protein and age, gender, smoking history, histological type, tumor size (P>0.05).Conclusion:1.The expression of Net-1mRNA and protein in NSCLC cancer tissues were both significantly higher than para-carcinoma tissues.2.The expression of Net-1mRNA is associated with TNM classification, differentiated degree, the higher grade malignancy of lung cancer, the higher expression of Net-1mRNA. The expression of Net-1protein in NSCLC cancer tissue is associated with TNM classification, differentiated degree, lymphatic metastasis.The higher degree of malignancy of lung cancer, the higher expression of Net-1protein.3.The change of expression in Net-1’s gene and protein levels may be related to advanced NSCLC.
