Background and objective:Migraine is a common and episodic primary headaches, but the pathology is poorly understood. Recent Genome-wide association studies (GWAS) have identified various migraine susceptibility variants. We aim to replicate3GWAS-associated polymorphisms (TRPM8rs10166942,LRP1rs11172113, MEF2D rs1050316) in Han Chinese of Hunan province.Methods:The study population consisted of migraineurs and healthy controls matched for age and sex. All clinical data were collected and the Genomic DNA was extracted from peripheral blood leukocytes of each subject by Phenol Chlorofrom Method. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis was used to analyze these SNPs (SNP rs10166942of TRPM8,SNP rs11172113of LRP1and SNP rs1050316of MEF2D).Frequencies of alleles and genotypes for the SNP rs10166942, SNP rs11172113, and SNP rs1050316were compared between migraineurs and controls. Clinical features were compared between different genotype groups.The data were analyzed by SPSS19.0. Results:1、135migraineurs and110controls participated in our study and both groups had no significant differences in sex and age.2、Genotype frequencies of three sites of SNPs were in Hardy-Weinberg equilibrium in patients and controls.3、No significant differences in allele and genotype frequencies of SNP rs10166942in TRPM8were observed between cases and controls.4、No significant differences in allele and genotype frequencies of SNP rs11172113in LRP1were observed between cases and controls.5、No significant differences in allele and genotype frequencies of SNP rs1050316in MEF2D were observed between cases and controls.6、No significant differences in onset of age, duration of disease, frequency of migraine, duration of attack, severity of migraine, aura and family history were seen in three SNPs of each wild group compared to mutant group.Conclusion:SNP rs10166942of TRPM8、SNP rs11172113of LRP1and SNP rs1050316of MEF2D with are not associated with migraine and clinical features of migraine in Han population in Hunan province.
