ObjectiveThis study was undertaken to screen the up-regulated lncRNAs and investigate the action of the lncRNA on osteoblast differentiation.MethodsFirstly, we cultured MC3T3-E1cells and induced differeciation. Then we extracted the general RNA in osteoblast differentiation and purified it, labeled cDNA and conducted PCR amplification. Fluorescence-labeled cRNA was purified and measured. The cRNA fragment was hybridized using microarray, and the up-regulated lncRNAs in osteogenic differentiation was screened. Then, qRT-PCR was conducted to verify the up-regulated lncRNAs in osteogenic differentiation. The expression of selected up-regulated lncRNA104was inhibited, and the production of OC and ALP were measured.Results19up-regulated lncRNAs were identified in osteogenic differentiation by microarray. The up-regulated expression of lncRNA642, lncRNA160and lncRNA104was confirmed by qRT-PCR in osteogenic differentiation, and was consistent with the results of microarray. The expression of lncRNA104was reduced to70%after the siRNA interference for48hours. The production of OC and ALP were inhibited after down-regulating lncRNA104.ConclusionWe have identified the up-regulated lncRNAs in mouse osteogenic cell differentiation. lncRNA104promotes the osteoblast differentiation.
