Objective:To investigate the expression of miR-21in COPD patients peripheral blood mononuclear cells and analyze the relationship between the expression of miR-21and serum concentration of IL-12p70in COPD.Methods:To select Peripheral blood mononuclear cells and peripheral blood specimens from four groups that met inclusion criteria between June and August2013:stable phase of COPD group(n=23), acute phase of COPD group(n=19), smoking group (n=19), health group (n=16)。Real-time RT-PCR was performed to determine the expression levels of miR-21of peripheral blood mononuclear cells of four groups; enzyme-linked immunosorbent (ELISA) was carried out to determine the concentration of IL-12p70in serum of four groups。To preliminarily explore the different expression levels of miR-21of peripheral blood mononuclear cells of four groups, and the correlation with concentration of IL-12p70in serum; detecting FEV1/expected value of stable phase of COPD group and acute phase of COPD group。Result:1.Compared with COPD group in stable phase,the expression levels of miR-21of peripheral blood mononuclear cells of COPD group in acute phase were elevated,but there was no statistically significant difference (p>0.05).the expression levels of miR-21of peripheral blood mononuclear cells of stable COPD group and acute COPD group are apparently higher than Smoking group and Health group (p<0.05),the expression levels of miR-21of peripheral blood mononuclear cells of smoking group are apparently higher than Health group (p<0.05).2.The concentration of IL-12p70in serum of acute COPD group is higher than other three groups (p<0.05),compared with stable COPD group and Smoking group and Health group.3.There was a positive correlation between the expression levels of miR-21of peripheral blood mononuclear cells and concentration of IL-12p70in serum of acute COPD group (r=0.341, p<0.05), similarly There was a positive correlation between the expression levels of miR-21of peripheral blood mononuclear cells and concentration of IL-12p70in serum of stable COPD group (r=0.306, p<0.05),There was no correlation between concentration of IL-12p70in serum and FEVl/expected value of acute and stable COPD group (r=-0.049, P>0.05; r=-0.073, P>0.05)。Conclusion:1.the expression levels of miR-21of peripheral blood mononuclear cells of COPD group were elevated, which is associated with lung function, miR-21may be closely related to the pathpgenesis of COPD.2.the concentration of IL-12p70in serum of acute COPD group is elevated,and there was a positive correlation between the expression levels of miR-21of peripheral blood mononuclear cells and concentration of IL-12p70in serum of acute COPD group,suggesting that miR-21may be involved in the regulation of IL-12p70expression.12figure,10table,44document.
