The Effect Of Downregulating Wnt-2Expression On The Proliferation And Growth Of MGC803

Objective：To analyze the influence of small interfering RNA targeting Wnt-2on the invasion,metastasis and proliferation of MGC-803gastric cancer cells.Preliminarily discussed thecorrelation between wnt-2gene and gastric cancer cell growth.Methods：On the basis of bioinformatics analysis,we chosed specific small interfering RNAtargeting wnt-2. The company constructed small interfering RNA targeting wnt-2.Then, wetransfected the siRNA targeting wnt-2into MGC-803cells through Lipofectamin2000,wedivided the cells into three groups:wnt-2siRNA group, scramble group and blank group..RT-PCR and Western blot were carried out respectively to observe the wnt-2expression ofMGC-803cells on the aspect of mRNA and protien. The cell scratch test was carried out toobserve the metastasis and proliferation ability of the cell. MTT was aimed to draw the cell,sgrowth curve.Results：1. The result of RT-PCR and Western blot showed that the expression of wnt-2decreasedmarkedly, the difference was significant(P<0.05).2. The result of Western blot showed that the expression of wnt-2decreased in the aspectof protien, the difference was significant(P<0.05).3.MTT result revealed that after48hours:wnt-2siRNA group’s OD(0.23±0.03)compared with scramble group(0.42±0.01) and blank group(0.47±0.03),wnt-2siRNA group was growing a little slower than the scramble group and blank group (P<0.05);72hours later, wnt-2siRNA group was growing obviously slower than scramble group and blank group(P<0.05),while there was no statistical differnce between scramble group and blank group (P>0.05).4.The cell scratch test exhibited: After48hours,the healing rate of MGC-803cells ofwnt-2siRNA group(18%±2.0%) was lower than scramble group(53%±2.7%)and blankgroup(41%±3.3%).72hours later, the difference was also significant,(P<0.05).Thehealing rate of MGC-803cells of wnt-2siRNA group was25%±2.5%scramble group was63%±2.6%; blank group was69%±3.3%.Conclusion：The MGC-803,s ability of proliferate and growth was significantly inhibited whenwnt-2was downregulated.
