| Objective: To explore the anti-anxiety and anti-depressant effects of hydrogensulfide (H2S) in streptozotocin(STZ)-diabetic rats and provide certain theorybasis for the development of a novel anti-depressant and anti-anxiety drug.Methods: Seventy-five adult male Sprague-Dawley(SD) rats, weight about300g, were divided into five groups randomly, that is control group, diabetesmellitus (DM) induced by streptozotocin (STZ) group, STZ+low concentrationof sodium hydrosulfide(NaHS)group, STZ+high concentration of NaHS groupand high pure concentration of NaHS group. STZ group, STZ+lowconcentration of sodium hydrosulfide(NaHS)group and STZ+high concentrationof NaHS group were performed by an intraperitoneal injection of STZ(40mg/kg).Venous blood was collected after three days. The diabetic model was built ifblood glucose concentration was exceed16.7mmol/L. While the control groupand high pure concentration of NaHS group were performed by anintraperitoneal injection of normal saline. Intraperitoneal injection of sodiumhydrosulfide(NaHS)were performed for STZ+low concentration of NaHS group,STZ+high concentration of NaHS group and high pure concentration of NaHSgroup, the dose is30μmol/d,100μmol/d and100μmol/d respectively for a month.All rats were performed four kinds of behavioral testing experiment such asspontaneous activity test(SAT), forced swimming test(FST), tail suspensiontest(TST) and elevated plus-maze test(EPM). FST, TST and EPM wereemployed for evaluating depressive-like and anxiety behaviors.Results:1) There were some changes about streptozotocin-diabetic ratsbehavior.①There is no significant difference between STZ group and controlgroup.②The immovable time of TST was significantly increased in STZ group than in control group.③The immovable time of FST was significantly increasedin STZ group than in control group.④The proportion of EPM entry open arm wassignificantly decreased in STZ group than in control group.⑤The time of EPMentry open arm was obviously shorter in STZ group than in control group.2)H2Shas several effect on STZ rats.①H2S has no significantly effect on the SAT ofSTZ rats.②H2S could shorten the Tail suspension test time of STZ rats.③H2Scould significantly shorten the Forced swimming test time.④H2S could increasethe proportion of EPM entry open arm in STZ rats.⑤H2S could significantlyincrease EPM entry open arm time in STZ rats.Conclusions①The STZ rats were in anxiety and depression state.②H2Scan relieve the depressive and anxiety behavior of streptozotocin-diabetic rats. |
