Objective:(1)To investigate the relationship between a-synuclein protein andP-4EBP1Protein in MPTP-induced parkinsonian mice.(2)To assess if P-4EBP1Protein and the TOR Signalling Pathway isinvolved in the processes of related-Parkinsonism proteins,a-synuclein’sexpression in MPTP-induced parkinsonian mice which was establishedwith Intraperitoneal injection MPTP.Methods:Fourty-eight parkinsonian mices model by MPTP directionalIntraperitoneal injection were equally randomized into4groups, Theparkinsonian mice model were Intraperitoneal injection by MPTP or SalineIV as design, Rapamycin was injectioned to the pretreating modle groupmice at the same time everyday in (7.5mg/kg)*7d. During the3-7th day,The parkinsonian mice model were Intraperitoneal injection by MPTP afterMedium injection30minutes;Medium was injectioned to the The modelgroup mice at the same time everyday inin (7.5mg/kg)*7d, During the3-7th day, The parkinsonian mice model were Intraperitoneal injection bySaline IV after Medium injection30minutes later;MPTP was injectioned to the The pretreating control group mice at the same time everyday inDuring the3-7th day in (7.5mg/kg)*7d.Medium was injectioned to theThe Blank control group mice at the same time everyday inin (7.5mg/kg)*7d.During the3-7th day, The parkinsonian mice model wereIntraperitoneal injection by Saline IV after Medium injection30minuteslater.The mice were observed behavior at same time point everyday.1hourafter the last LPS injection,Kill the parkinsonian mice and brain tissue ofmice Brain tissue was taken out observe the P-4EBP1and a-synucleinexpression in mice substantia nigra and striatum byimmunofluorescence.observe the attachment of SN stained cells,Qualitative and quantitative detection of the relationship between theexpression of a-synuclein protein and P-4EBP1Protein byWB.Analysis:①To investigate the relationship between a-synuclein proteinand P-4EBP1Protein in substantia nigra and striatum of MPTP-inducedparkinsonian mice.②To assess if P-4EBP1Protein and the TOR SignallingPathway is involved in the processes of related-Parkinsonism proteins,inMPTP-induced parkinsonian mice which was established withIntraperitoneal injection MPTP.Results(1)the effects of Behaviors Changes was higher in the pretreatingmodel group than in the model group and the Blank control group is muchbetter thanThe pretreating control group; (2)The results of immunofluorescence staining:the expression ofP-4EBP1and α-synucleinin Protein in the model and pretreating group wasbetter than the control group(p<0.05);the expression of P-4EBP1andα-synucleinin Protein in the Intervention group was lower than modelgroup。(3)The results of Western blote:the expression of P-4EBP1andα-synucleinin Protein the model group is better than the blank controlgroup(p<0.01);the expression of P-4EBP1and α-synucleinin Protein in thepretreating model group was lower than model group(p<0.01)。Conclusions:(1)The experimental results:P-4EBP1is involved in the pathogenesisof PD through TOR signaling pathway in parkinsonian mices model byMPTP directional pretreating injection(2)TOR inhibitors is affected by the negative regulation of P-4EBP1expression,Reduce the expression of α-synuclein in parkinsonian micesmodel,chang the beahaver of parkinsonian mices model.(3)P-4EBP1may be a potential target for the clinical treatment of PD,rapamycin may have potential effectfor clinical PD.
