Objective:This study was designed to explore:①The cell viability and proliferation of human non-small cell lung cancer cell line A549and H460,which treated on ginsenoside Rd(12.5-100μmol/L)in vitro;②The effect of ginsenoside Rd in vitro on the invasion capability of A549and H460cells;③The effect of ginsenoside Rd on the cell cycle of A549and H460cells;④To detect the expression of cyclin Dl,cyclin El,E2F1,CDK2,CDK4,p27,p21and pAkt by western blot analysis.Method:(DMTT method was used to observe the effects of different concentrations of ginsenoside on human non-small cell lung cancer line A549and H460proliferation and the relationship between drug effect and time;②Detect the effect of apoptosis of ginsenoside Rd by high content screening;③Transwell invasion assay was used to observe the invasion capability;④Flow cytometry, method was used to observe the cell cycle induced by different concentrations of ginsenoside Rd in human non-small cell lung cancer line A549and H460;⑤Expression of these proteins which are important in cell cycle G1phase to S phase node were measured by western blot.Resμlts:①ginsenoside Rd inhibited the proliferation of human non-small cell lung cancer cell line A549and H460with a certain degree of concentration-dependence:it significantly inhibited the proliferation when the drug concentration increased to100μmol/L and the inhibition gradually increased with time.②Ginsenoside Rd applied on human non-small cell lung cancer A549and H460cells, induced the cell apoptosis with a certain degree of concentration-time dependence.③The invasion capability of A549and H460cells decreased after treatment of ginsenoside Rd.④As compared with the negative control group, the percentage of the cells in G0/G1phase increased in treatment group, while the percentage of the cells in S phase decreased, which showed the cells were arrested at the G0/G1phase of the cell cycle.⑤Western blot detection the expression of cyclin D1, cyclin E1, E2F1, CDK2/4, pAkt and p27were decreased with increasing concentration of ginsenoside Rd, while the increased expression of p21.Conclusions:①Ginsenoside Rd inhibited the proliferation of human non-small cell lung cancer cell line A549and H460.②Ginsenoside Rd suppresses human non-small cell lung cancer cell line A549and H460invasion capability.③Ginsenoside Rd can hinder the cell cycle at G1phase.
