Study On Promoting Blood Circulation And Chemical Constituents Of Dracocephalum Moldavica L. Xin Jiang

Objective:The subject of{Dracocephalum moldavuca L.) anti-platelet aggregation role of the active ingredient and to study the antithrombotic role, according to the results of activity tracking, separation and purification of active fractions and obtained active monomer composition, to further explore the monomer composition of anti-platelet aggregation, to provide for the development of medicinal materials and their preparation basic information. Methods:1. To determine the effect of Dracocephalum moldavuca L. on platelet aggregation, we observed two kinds of extraction site, three kinds of extraction site, six kinds of monomer compounds on platelet aggregation effect; using turbidimetry to discuss what active components to able to have the anti-platelet aggregation function.2. Uses the reflux method, the water,95%ethanol in turn carries on the extraction to the Dracocephalum moldavuca L. dry aerial parts, water extracts respectively extraction by chloroform, ethyl acetate, n-butanol and the extracts were isolated and purified using silica gel column the macroporous resin, and the structures were identified by the data of UV> IR、1H-NMR、13C-NMR.3. Two kinds of experimental models in vivo, Pulmonary thromboembolim in mice and ligation-induced inferior caval thrombosis in rat were used to examine the antithrombotic activity of aqueous and alcohol extract of Dracocephalum moldavica L. Results:1. In vitro results show that Dracocephalum moldavica L. alcohol extracts, chloroform, ethylacetate, n-butanol fractions have effectively inhibit platelet aggregation induced by thrombin; acacetin and tilianin have strong anti-platelet aggregation.2. Dracocephalum moldavica L. different extracts of100mg/kg and400mg/kg can markedly increase the survival rate of the mouse suffering from Pulmonary thrombosis induced by the mixture of collagen and epinephrine (P<0.05or P<0.01) and significantly decreased the weight of thrombus ligation of inferior vena cava (P<0.05or P<0.01); the dose were400mg/kg alcohol extracts effect is most significant (P<0.01).3. Sewen compounds were isolated from, Dracocephalum moldavica L. they are acacetin (Ⅰ),tilianin (Ⅱ),p-hy-droxycinnamic acid (Ⅲ),methyl p-hydroxycinnamate (Ⅳ), Daucosterol (V), β-Sitosterol (Ⅵ). Conclusion:This research through the Dracocephalum moldavica L. extracts、different fractions and chemical compounds anti-platelet agglutinating and antithrombotic study, in order to associate promoting blood circulation to remove blood stasis role of material foundation, and supplied data information for development of medicinal materials and preparation of Dracocephalum moldavica L.
