Objective:Establishing the rat model of severe acute pancreatitis, rats in treatment group were administrated with single rhubarb, detecting and comparing the mRNA expression level of Toll like receptor2、4on intestinal mucosa in each group to make sure whether rhubarb can decrease the mRNA expression level of Toll like receptor2、4on intestinal mucosa and protect the intestinal mucosal barrier, thus inhibiting inflammatory reaction and controling the development of severe acute pancreatitis effectively.Methods:Totally24rats were divided into sham operation group, pancreatitis group and treatment group respectively. The SAP model was built with retrograde injection5%sodium taurocholate through biliopancreatic duct. The treatment group was treated with10%rhubarb soup. The blood samples were collected for measurement of serum amylase; histological changes of pancreas and terminal ileum were evaluated by the corresponding score system;the changes of expression level of TLR2、TLR4mRNA were detected by reverse transcription polymerase chain reaction in intestinal tissues.Result:The TLR2、TLR4mRNA expression level of the pancreatits group were higher than those of sham operation group, and those of treatment group were lower than those of pancreatitis, the difference was significant (P<0.05) respectively. Conclusion:Rhubarb can reduce the mRNA expression level of TLR2、TLR4in severe acute pancreatitis and protect the intestinal mucosal barrier, thus inhibiting the further development of inflammatory reaction and decrease the injury of rats pancreas.
