| Objective:To investigate the application prospect of dinucleotide STR selected in paternity testing. Methods:Dinucleotide STR loci D6S261ã€D15S216were selected,200unrelated blood samples from Shanghai were collected,to investigate the allele frequency and statistical parameters for forensic idengtification.and193paternity testing blood samples including58normal samplesã€9mutation samples were collected to investigate the application of dinucleotide STR in paternity testing.The touch-down PCR condition was used,and declined the denaturalization temperature to85℃,then amplified.the PCR products were collected by3130x1Gene-tic Analyzer,and analyzed with PowerStatsv12.xls. Results:15alleles,50genotype and Hã€DPã€PEã€PIC were found from200unrelated blood samples of D6S261;mutation were not found in193paternity testing blood samples;Only6alleles were found in D15S216,then delected D15S216.Conclusion:D6S261is highly polymorphic, decrease more stutter bands, the characteristic of dinucleotide STR with low mut-ation rate and high stab-ility,is a effective method to solve the indetermination of paternity testing caused by mutation. |
