Background:The miRNAs are a class of18-25nt non-protein coding RNA, play a role in inhibiting the translation of target genes.miRNA effect the tumor occurrence.The miRNA is currently a hot research, may become the target of the cancer diagnosis and treatment.Osteosarcoma, the most common malignant primary bone tumor, occurs in young man which is fast-growing, short course, high mortality in the clinical.Now the diagnosis of osteosarcoma mainly relies on clinical symptoms, imaging studies plus pathological diagnosis based on a model. Now we lack of a kind of early clinical, noninvasive, sensitive biological markers. This study was designed to the expression of miRNAs in the serum of osteosarcoma.The signature could use to the diagnosis and the target treatmentObjective:To detect the miRNA expression level in the serum of osteosarcoma patients, find the MiRNA expression signature in the serum of osteosarcoma.To find the further study of osteosarcoma and miRNA diagnosis and treatment. Materials and methods:Use the microarray to find the signature of miRNA expression profiles in the serum of patients of osteosarcoma. and apply real-time fluorescence quantitative PCR(stem-loop RT-qPCR, the TaqMan probe method) validation expression profiles in the serum of osteosarcoma. Collection of patients with osteosarcoma serum10cases and10cases of normal, healthy human serum, use the array to the initial detection of serum miRNA differences in expression profiles of microarray data using SAM system statistics. And choose U6snRNA as endogenous control, and qRT-PCR measurements to detect the results in relatively normal population is upregulated and downward adjustment of miR-199a-5p, miR-199a-3p, miR-133a and miR-515-5pResults:1.The miRNA biological microarray and RT-PCR can be used to detect the miRNAs in the serum of patients with osteosarcoma and serum of normal healthy people.2.Using the micro array can detect the series of miRNA difference expression in patients with osteosarcoma and serum of healthy people.3.The qRT-PCR could dectect miR-199a-5p, miR-199a-3p, miR-133a and miR-515-5p in the serum.And the results are the same as the array method. Conclusion:1.There are miRNAs in human serum,which can be detected convenietly bymiRNA biological microarray and RT-PCR.2. The array could detect the significant difference signature between the group of osteosarcoma patients and the healthy people.And the RT-PCR validation of miR-199a-5p, miR-199a-3p, miR-133a and miR-515-5p can be the candidate serum markers of osteosarcoma13.There may be specific serum miRNA expression signature.
