Objective The study was designed to investigate what compounds were absorbed in rat brain with Traumatic Brain Injury (TBI) following oral Danchuanhong (DCH), to compare the anti-oxidation of absorbed compounds with the DCH and to explore the material basis for the efficacy of DCH in protection against TBI via the anti-oxidation pathways.Methods The Ultra-high Performance Liquid Chromatography with Tandem mass spectrometry (UPLC-MS/MS) was used to detected the absorbed compounds(Tanshinol, T; Ferulic acid, FA; or Hydroxysafflor yellow A, HSYA) in brain following oral administration of DCH about30min to TBI rats. Then72Sprague-Dawley rats with TBI were randomly divided into sham TBI, TBI+Vehicle, TBI+DCH and TBI+HSYA groups. The samples of cerebral cortex in each group were collected at6,12and24hour after TBI and analysed for their oxidant-antioxidant status via detecting the level of malondialdehyde (MDA), the activities of superoxide dismutase (SOD), catalase (CAT), and the contents of glutathione (GSH), GSH/GSSG ratio. Results1. Success to establish the UPLC-MS/MS method for detecting the absorbed compounds following gavage of DCH.2. HSYA (m/z,611→491.04or611→325.06) and FA(m/z192.8→177.95;192.8→133.96) in water extract of DCH was determined in sample of brain tissue.2. DCH and HSYA group dramaticlly reduced the level of MDA, increased the activities of SOD, CAT, and the contents of GSH, GSH/GSSG ratio in cerebral cortex with injury (p<0.05). There was no statistically significant difference compared each other (p>0.05).Conclusion HSYA exerts anti-oxidation similar to DCH in protection against TBI.
