Purpose:Combing and summarize the ancient and modern physicians’ understanding of etiology and pathogenesis, therapeutic principles of primary osteoporosis comprehensivly, through the systematic arrangement and research on the compatibility method of the effective prescriptions in the ancient and modern literature about the treatment of primary osteoporosis, explore the correlation of drug compatibility and regularity of primary osteoporosis of ancient and modern physicians. Observe classical reinforcing-kidney prescription-Zuogui Wan and Yougui Wan’ intervention of the primary osteoporosis model, reveal the reinforcing-kidney method, efficacy of the treatment of primary osteoporosis and some of the mechanisms, in order to provide reference for in-depth study of traditional chinese medicine for osteoporosis’prevention and cure function.Methods:1.The theory disscution:According to the literature of the Qin and Han dynasties to Qing dynasty more than seventy ancient medical books and publishd since1990in the journal of Chinese medicine on primary osteoporosis etiology and pathogenesis and treatment,explore the understanding and developing procedure of OP in Chinese medicine. Summarize the origin and development for the physicians’ using reinforcing-kidney method for the OP treatment.2.Statistical analysis:Collect a total of114prescriptions in ancient and a total of308prescriptions in modern to establish an effective data base,use Rapidminer software to explore the statistical frequency and correlation to find the compatibility of reinforcing-kidney drugs in the Chinese medicine treatment on primary osteoporosis.3.The animal experiment:Through the copy of primary osteoporosis rats which were excised the ovaries model,observe the classical prescription Zuogui Wan and Yougui Wan to intervene the model right from the pathological morphology, bone mineral density and serum bone metabolism biochemical indicators.Results:1.The origin analsis:Traditional Chinese medicine has a long history in the treatment of OP,the doctors of Qin and Han dynasties founded the theory of kidney advocating bone,lay the method of reinforcing-kidney to strengthen bone; The doctors of Sui and Tang dynasties put forward the theory of raising-liver to strengthen bone,exploit the idea of strengthening Jing and Blood;The Song and Yuan dynasties put forward the theory of tonifying-spleen to raise marrow and the prevention and treatment of OP;The doctors of Ming and Qing dynasties inherited the experience of predecessors,lay attention to invigorate the blood circulation and expel the pathogen;The modern doctors made the diagnostic criteria and preliminary build the syndrome differentiation system. The treatment of strengthening-kidney run through the development of the treatment on OP.It is the fundamental method of solution for OP.2.The commonly used drugs:The ancient prescription drugs have been used for1266times,involving162kinds, the modern prescription drugs have been used for3090times, involving172kinds of drug. They are concentrated in the flowing aspects such as tonifying-deficiency, promoting blood circulation to remove blood stasis,warming-interior, removing the water and moisture, dispelling wind and dampness, astring, clearing heat, etc.Among the ancient prescriptions,the top three are tonifying-deficiency medicine accounted for43.29%,warming-drugs accounted for9.56%, promoting blood circulation to remove blood stasis drugs accounted for7.66%. Among the modern prescriptions,the top three are tonifying-deficiency medicine accounted for59.00%, promoting blood circulation to remove blood stasis drugs accounted for15.21%,medicine for rheumatism accounted for5.24%.Among them,the reinforing-kidney and invigorating-essence drug have a heavy proportion.The drugs with high frequency are Shudihuang, Duzhong, Yinyanghuo, Lujiao,Roucongrong, Tusizi, Buguzhi,Bajitian,Chuanduan,etc;Invigorating spleen and supplementing qi are also important,such as Huangqi,Renshen,Shanyao,Baizhu, Dangshen;Nourishing blood and yin of liver and kidney drugs,such as Gouqizi,Guiban,Nvzhenzi,Danggui are used frequently;Among the promoting blood drugs,Niuxi,Chuanxiong,Gusuibu and Danshen are mostly used;Among the warming interior drugs,Fuzi and Rougui are commonly used;Among the removing wind-dampness drugs,Gouji,Sangjisheng,Duhuo and Weilingxian are mostly used.3.The correlation analysis:In the ancient prescriptions,Duzhong and Niuxi,Shudihuang and Niuxi,Rougui and Shudihuang,Duzhong Rougui and Duzhong and Niuxi and so on appear frequently. Most of them are combinated by Shudihuang and Niuxi as the latter rule.In the modern prescriptions,Yinyanghuo and Shudihuang,Duzhong and Shudihuang, Huang qi and Yinyanghuo,Yinyanghuo Danggui and Shudihuang,Shanyao Shanzhuyu and Shudihuang and so on are frequently appear. Most of them are combinated by Shudihuang, Yinyanghuo and Shanyao as the latter rule.In the ancient and modern prescriptions,Shudihuang appeared most as the core with high frequency. The high frequency drugs in the association rules such as Dihuang,Duzhong,Danggui,Shanyao,Shanzhuyu,Niuxi,Lujiao,Tusizi,Fuling,Chuanduan,Gouqizi, Guiban,Renshen,Fuzi,Rougui,Huangqi,Yinyanghuo can be regarded as an adding and subtracting changes from Liuwei Dihuang Wan,Zuogui Wan and Yougui Wan.4. Experimental observation:By copy of primary osteoporosis rats which were excised the ovaries model, take the classical prescriptions Zuogui Wan and Yougui Wan to intervene, found that Reinforcing-kidney method can improve pathological morphology of the rat model, increase bone density, improve serum bone metabolic biochemical indicators.Conclusions:1.Traditional Chinese medicine has a long history in the treatment of osteoporosis and has accumulated rich experience.Give priority to reinforcing-kidney to strengthen bone,accompanied with raising-liver,tonifying-spleen and invigorating the blood circulation along with the changes of syndrome. It is the treatment characteristics of primary osteoporosis.2.In the ancient and modern physicians, Shudihuang,Niuxi, Yinyanghuo and Shanyao are the main composition.The compatibility method embodies the adding and subtracting changes from Liuwei Dihuang Wan,Zuogui Wan and Yougui Wan.3. The reinforcing-kidney prescriptions Zuogui Wan and Yougui Wan are effective in the treatment of osteoporosis from pathological morphology,bone mineral density and serum bone metabolism indexs.The mechanism may be through inhibiting the high conversion state of castration rats.
