| Objectives: To investigate the inhibition effect of Diallyl disulfide (DADS)on proliferation of rat hepatic stellate cell and secretion of extracellularmatrix,and to explore whether DADS can inhibit the Wnt/β-catenin signalingpathway by increasing retinoic acid receptor-related solitary nucleus α (RORα)to block the progress of liver fibrosis.Methods: HSC-T6cells were treated with different concentrations of DADS(10ã€20ã€40ã€80)mg/L for different period time, and then the cell prolifer-ation wasexamined by MTT.The expressions of liver fibrosis â…¢ collagen (PCâ…¢),laminin (LN)were assessed by ELISA. The mRNA and protein expression of β-catenin, cyclin D1and RORα were examined by RT-PCR and Western Blot respectively.Results: MTT test indicated that DADS inhibited the HSC-T6proliferation in adose-dependent manner with its most obvious effect forty eight hours afterintervention and the IC50of42.06mg/L. DADS reduced the expression of PCâ…¢, LNsignificantly in the ELISA assay (P<0.05). RT-PCR test indicated that the mRNAexpression of β-catenin and cyclin D1were inhibited by DADS (P<0.05).Western Blot results showed that DADS can down-regulate the protein expressionof β-catenin, cyclin D1and up-regulate RORα expression (P<0.05).Conclusion: DADS may suppress HSC-T6cell proliferation and secretion ofextracellular matrix by inhibiting the Wnt/β-catenin signaling pathway by wayof up-regulating the expression of RORα. |
