Objective To investigate the protective effect of high mobility groupbox(lHMGBl)gene silence on oxygen-glucose deprivation/reoxygenation(OGD/R)-induced injury in astrocytes in vitro.Methods Astrocytes were divided into control group,OGD/R groupand HMGB1siRNA group.Astrocytes in OGD/R group were treated withOGD for2,4,and6h respectively,and then reoxygenated for24h.InHMGB1siRNA group, astrocytes that transfected with HMGB1smallinterference RNA(siRNA) lentivirus-vector were treated with OGD6h,and then reoxygenated for24h.The protein of normal astrocytestransfected with HMGB1small interference RNA (siRNA)lentivirus-vector were inspected by Western-blot.After24hours’reoxygenation,The mRNA and protein expression of HMGB1in theastrocytes were detected by RT-PCR and ELISA method.The cell injuryand survival rate were inspected by LDH activity(LDH%) and MTTassay. Results Compared astrocytes without transfected with HMGB1small interference RNA(siRNA)lentivirus-vector,The protein expression of HMGB1can be suppressed by small interference RNA. Compared withnormal group,as the OGD time prolonged,the mRNA and proteinexpression of HMGB1were increased gradually(P<0.05) after OGD/R,which were further increase with time elapse. OGD/R resulted insignificant injuries with time prolonged,the LDH%was increased(P<0.05) and the survival rate was decreased severely(P<0.05).Compared with OGD/R group,cell injury in HMGB1siRNAgroup was remarkably decreased,as survival rate was raised significantly.Conclusions Expression of HMGB1in astrocytes can be inhibitedby small interference RNA,over-expression of HMGB1might be animportant factor in OGD/R-induced cell injury.... |
