| Objective:To research the change of the acute toxicity and pharmacological effects before and after processed in Aconitum leucostomum Worosch, and compared it with radix Aconiti kusnezoffii who has have quality control already, and to lay a good foundation of its clinical application and the further study of the pharmacodynamics of Aconitum leucostomum Worosch and its processed products. Methods:1. Using the method of HPLC to determine content of Aconitum lcucostomum Worosch’s water extracted solution.2. Using the method of intraperitoneal injection of mice to determine LD50of Aconitum leucostomum Worosch and its processed products.3. Taking radiant heat stimulation and acetic acid induced writhing test to investigate the analgesic function of Aconitum leucostomum Worosch and its processed products.4. To observe the anti-itnflammatory effects of Aconitum leucostomum Worosch and its processed products to CIA rats. Results:1. The content order of Aconitum leucostomum Worosch and its processed products: Aconitum leucostomum>Kazakh method>Pharmacopoeia method (2010)>Literature method.2.(1)The toxicity order of Aconitum leucostomum Worosch and its processed products:Aconitum leucostomum>Pharmacopoeia method(2010)>Kazakh method>radix aconiti kusnezoffii>prepare radix aconiti kusnezoffii>L>terature method; The result showed that the LD50were1.848g/kg,4.742g/kg,5.846g/kg,6.139g/kg,19.795g/kg,27.172g/kg respectively.(2)Within1~2h after intraperitoneal injection of mice, a portion of mice in each group appear the poisoning symptoms of every system, especially on the central nervous system, cardiovascular system, digestive system and so on.3.(1)Within0~120min, the pain threshold of each group of mice who suffered with radiant heat stimulation, all present this trend:first increases and then decreases, while60min, the pain threshold of mice reached the maximum.(2)Compared with the control group, in addition to pharmacopoeia method, Kazakh processed products’ low dose group, literature processed products’ low and middle dose group have weak analgesic effect, the rest of the group could significantly prolong flick latencies of mice caused by the radiant heat stimulation in mice (P<0.01or P<0.05), to have analgesia effect.(3)Compared with the control group, in addition to Aconitum leucostomum, its pharmacopoeia method, Kazakh processed products’ low dose group, literature processed products’ low and middle dose group, the rest of the group can reduce the times of body torsion of mice (P<0.01or P<0.05), to have analgesia effect.4.(1)Aconitum leucostomum and its processed products could significantly inhibit joint swelling rate of CIA rats in different degrees.(2)When Aconitum leucostomum and its processed products observed under light microscope, each medicated group can remarkable improve the pathological feature of its synovial tissue. The improvement of the pathological feature of its synovial tissue by Aconitum leucostomum and its processed products is better than Radix Aconiti Kusnezoffii and its processed product.(3)6weeks after the treatment, in addition to the low dose group of Aconitum leucostomum and its processed products, the rest of each group’s SA, IL-2and TNF-a level in serum of CIA rats is lower than the model group (P<0.01or P<0.05).Conclusion:1. The content, acute toxicity and pharmacodynamics experiment show that the content of Aconitum leucostomum’s pharmacopoeia and Kazakh method processed products were better than its literature method processed products, the toxicity of Kazakh method processed products is smaller than pharmacopoeia method, and its analgesic and anti-inflammatory effect is better than the pharmacopoeia and literature method processed products.2. Compared with Radix Aconiti Kusnezoffii, in addition to the literature method processed products, the toxicity of Aconitum leucostomum and its processed products were greater than Radix Aconiti Kusnezoffii and its processed products, but its effects of analgesic and anti-inflammatory of mice were better than Radix Aconiti Kusnezoffii and its processed products.3. Aconitum leucostomum can reduce the poison effect after processing, has a certain medicine value. In the aspect of acute toxicity and pharmacologic actions, its Kazakh method processed products is better than the other two kinds of processing methods, so we should priority take the Kazak method processing methods as Aconitum leucostomum’s best processing technology to reduce toxicity and maintain efficacy. |
