| ObjectiveTo analysis of Flos Albiziae active ingredients and compare of the effect ofsedative and hypnotic of Flos Albiziae and the fruit of Celastrus orbiculatus.Studied of Flos Albiziae and the fruit of Celastrus orbiculatus on the effect oncerebral cortical neurons of insomnic rats, and to explore their mechanism ofaction.MethodsTo establish a method for the simultaneous determination ofisoquercitrin,quercitrin and quercetin in Flos Albiziae. Optimizing the extractionrate of total flavonoids in Flos Albiziae by using the Single factor experimentand orthogonal experiment, in order to determine the extraction method.Pharmacodynamics mainly consists of ethology aspect and histology aspect.The ethology part, comparison by using the extractions of Flos Albiziae and thefruit of Celastrus orbiculatus on the spontaneous activity of rats, and theinfluence on the sleep induced by pentobarbital sodium. Using HE staining andthe S-P method for immunohistochemical staining to observe the structure ofcells in cerebral cortex and the expression of5-HT and GABA.ResultsThe influence factors of the extraction of total flavonoids from Flos Albiziaewere reflux-temperature, ethanol concentration, the ratio of material liquid andreflux-time. The best extraction process was90%ethanol,1:25of the materialliquid, reflux-time was1h, reflux-temperature was90℃. On the condition, theextraction of total flavonoids from Flos Albiziae was2.41%. The peak area andthe concentration showed a good linear relationships of isoquercitrin,quercitrinand quercetin were7.12-356μg·mL-1(r=0.9998,n=6),12.12-606μg·mL-1 (r=0.9998,n=6),5.28-264μg·mL-(1r=0.9997,n=6),respectively. The ethylacetate and n-butanol extraction group of Flos Albiziae significantly reducedthe spontaneous activity of rats, increased the number of subthreshold dose ofpentobarbital sodium induced sleeping number and collaborate threshold doseof pentobarbital sodium, prolonged the sleeping time of rats. To observe thestructure changes of nerve cells under the light microscopy, the ethyl acetateand n-butanol extraction group of Flos Albiziae significantly less than modelgroup, and not significantly different from the normal group. The results ofimmunohistochemistry showed that the expressions of5-HT and GABAsignificantly increased in the ethyl acetate and n-butanol extraction group ofFlos Albiziae compared with the model group. But the difference was notsignificant between the other treatment group and the model group.ConclusionsResearch showed that the best solution for extraction of total flavonoids ofeffective components of Flos Albiziae is1:25,90%ethanol, reflux-time was1h,reflux-temperature was90℃, the ratio of extraction was2.41%. The methodwas reproducible, simple and reliable. Using the RP-HPLC to determinate thecontent of isoquercitrin, quercitrin and quercetin in Flos Albiziae, the methodwas sensitive, accurate, reproducible. The sedative and hypnotic effect of FlosAlbiziae was significantly better than the fruit of Celastrus orbiculatus, and thepart of ethyl acetate was the best. And its mechanism may be related to protectthe cell structure normal and enhance the expression of5-HT and GABA. Inaddition, the sedative and hypnotic effect of the fruit of Celastrus orbiculatuswas not significant. |
