| Objective:To study the expression differences and characteristics about different su btypes of breast cancer in PI3K/Akt/mTOR signaling pathway proteins,in order to e xplore this pathway in occurring and development in triple negative breast cancer.Methods:Selected117cases which were diagnosed in invasive breast cancer, whic h come from January2005to June2010and Cancer Hospital of Xinjiang Medical University,according to the result of immunohistochemical of molecular types abou t ER/PR and HER-2,we divided them to two groups,that is triple-negative breast ca ncer group(TN group,28cases),non-triple-negative breast cancer group(non-TN group,89cases).Then by using SP method to detection the expression of associated protei n and its phosphorylation about PI3K/Akt/mTOR in breast cancer and its expressio n in cancer tissues adjacent,in order to analyze the differences of expression and c haracteristics in different types of breast cancer organizations,and to analyze the cor relation in the clinical pathological features.Results:(1)The expression rate of associated protein and its phosphorylation about PI3K/Akt/mTOR in breast cancer tissue is higher,both P<0.05.(2)The characteristics of activation in different subtypes of breast cancer tissues of PI3K/Akt/mTOR sig naling pathway,the positive expression rate of EGFR in triple negative breast cance r was significantly increased,while PI3K,p-PI3K,Akt and p-S6K1in non-triple-negati ve breast cancer positive rate increased significantly.At the same time,there are diff erences between the protein phosphorylation,mostly showed significant positive corr elation.(3) The expression of EGFR activated PI3K/Akt/mTOR pathway,there are si gnificant positive correlation expression when EGFR positive expression of PI3K a nd p-PI3K,Akt and p-Akt473,p-mTOR and p-S6K1.(4) The downstream PI3K/Akt/m TOR signal pathway of p-mTOR,p-S6K1and p-4EBP1were associated with the tu mor size,Status of axillary lymph node and TNM stage,suggesting the clinical signi ficance of triple-negative breast cancer and non-triple-negative breast cancer progno sis.Conclusion:The signaling pathway about PI3K/Akt/mTOR is obviously activated in the conclusion of breast cancer;the expression is different in signaling pathway of associated protein about PI3K/Akt/mTOR,that means we can make different indivi dual strategy according to different subtypes of breast cancer. |
