Objective:To investigate the association of the single nucleotide polymorphisms (SNPs) at the gene for tumor necrosis factor-a (TNF-a)(rs1799964) with the variable susceptibility to Uygur patients with pigeon breeder’s lung. Methods:The data and blood samples were enrolled as described (92subjects including62feeding pigeons and30normal control group).DNA was isolated from blood samples.Primer design, sample preparation,PCR optimization,SAP and single base extension were performed by using MassARRAY technology according to SNP loci appendix sequence information,SNPs at the genes for TNF-a(rs1799964) was analyzed using mass spectrometry.Statistical analyses were performed by with SPSS17.0statistical software,genotypes and allele frequencies at each SNP were compared by using the chi-square test,and calculated the relative risk value(OR). Results:1.subjects with the T/T or T/C or C/C genotype at TNF-α-1031T→C were found,which were tested for Hardy-Weinberg equilibrium (HWE).2.There were significant differences in the distribution of T/T or T/C or C/C at TNF-α(rs1799964) among three groups(x2=9.679, P=0.046),no difference found in the distribution of T/T or T/C or C/C comparing case group and negative control group and normal control group (x2=3.865,P=0.145,x2=2.94,P=0.23);while Subjects with G/G or G/T or T/T,TNF-a showed a significant difference in the distribution of between negative control group and normal control group(x2=5.455,P=0.02);3.The frequency distribution of allele T in patients with PBL was higher than in the negative control group lower than in the normal control group,the frequency distribution of allele C in patients with PBL was lower than in the negative control group, higher than in the normal control group,no difference found in three groups(x2=4.755,P=0.093), there were no differences in allele between case group and negative control group or normal control group (x2=0.162,P=0.687, x2=3.224,P=0.073);there was significant difference in allele comparing negative control group or normal control group (x2=4.615,P=0.032). Conclusion:1.SNPs at the gene for TNF-a (rs1799964) was associated with the variable susceptibility to Uygur patients.2.Not found the TNF-a (rs1799964) gene polymorphism is associated with PBL susceptibility, further studies are warranted to verify our results.
