Objective To investigate the correlation between the chronic periodontitis andMonocyte chemoattractant protein(MCP) and the role of MCP-1in pathogenesis ofchronic periodontitis, we extracted gingival crevicular fluid before and after periodontalnon-surgical therapy in patients with chronic periodontitis, the change of theconcentration of MCP-1and observed the relationship between the groups. The changesof MCP-1in gingival crevicular fliud after periodontal therapy in periodontitis patientsrevealed, in this study provide basis for the link between cardiovascular disease andperiodontitis, and some guidance on the prevention and clinical treatment.Methods Subjects seleted:40patients were seleted (20people of each gender and agevarying from25to60years old) from2012to2013in dental outpatient department inGeneral Coal Hospital,20of them with chronic periondontitis and20as healthy control.Inclusion criteria: tooth with complete shape,tooth neck without decay, without fillings,no large tartar, gingival index>1, attachment loss≥3mm, or probing depth≥4mm.Exclusion criteria: systemic diseases such as hypertension, diabetes, inflammatorydiseases, pulmonary infection, virus or fungal infections, rheumatoid arthritis andrecently receiving periodontal treatment, using non-steroidal anti-inflammatory drugs andother medical history in recent3months, history of smoking. The testing method:1Before the trial,sufficient communication with the patients is needed, in a bid to to obtainthe permission to divide into groups.2We choose moisture absorption paper pointmethod to extract gingival crevicular fluid from the most serious part of the periodontitisof the subjects, and make records. All operations are performed by the same experimenterand extracting gingival crevicular fluid from the same place.3Put the finished samplesimmediately into the prepared EP tubes and stored in-70℃low temperature refrigerator.4Recording clinical test indicators, including plaque index, bleeding index, loss ofattachment and probing depth.5Experimental subjects receive periodontal non-surgicaltherapy including oral hygiene education, plaque control, supragingival scaling,subgingival scaling, root planing and occlusal adjustment, etc.6To detect the collectedsamples by enzyme linked immunosorbent test (ELISA),and get the data.7Using SPSSsoftware for data analysis, getting and analyzing the result. Results The concentration level of MCP-1in gingival crevicular fluid of chronicperiodontitis group is significantly higher than the control group and that of chronicperiodontitis group after periodontal non-surgical therapy is significantly lower thanbefore, which shows significantly statistical differences (P<0.05), while there was nostatistically significant difference in MCP-1level between chronic periodontitis groupafter treatment and control group(P>0.05). Clinical indicators such as gingival index (GI),plaque index (PLI), probing depth (PD), attachment loss (AL) and bleeding index (BI)show significantly statistical difference (P<0.05) between the chronic periodontitis groupbefore the therapy and control group. The clinical indicators of chronic periodontitisgroup before and after periodontal non-surgical therapy showed significantly statisticaldifference(P<0.05).Conclusion The concentration of MCP-1of gingival crevicular fluid in patients withchronic periodontitis is significantly higher than that of healthy subjects and significantlydecreased after periodontal non-surgical treatment, so as to as the clinical indicators suchas plaque index, gingival index, attachment loss, probing depth,et al. The variation ofconcentration of MCP-1can reflect the severity and progress of chronic periodontitis,which can be used as an index of judging clinical stage of chronic periodontitis andprovide certain basis for the prevention and treatment of chronic periodontitis.
