| Objective To observe the effect of Dong qin qing jin mixture on the cell proliferationand expression of pro-collagen in MRC-5human embryonic lung fibroblast (HELF)irradiated by X-ray, and to provide some new ideas and experimental basis for screeninganti-radioactive pulmonary fibrosis (RPF) drugs.Methods1The cell model was made and study the dose-and time manner of X-rayirradiation in HELF:1) dose manner: MRC-5cells was irradiated with X-ray for5,10,12,15,18,20Gy doses. The cell proliferation was measured by MTT assay in48h afterirradiation.2) time manner: MRC-5cells was irradiated with X-ray in a5Gy dose for24,48and72h, then the total proteins were extracted and the expression of pro-collagentype I and III were measured by western blot.2. The anti-radioactive fibrosis effect ofDong qin qing jin mixture: MRC-5cells were divided into5group, just as1) blank group(group C);2)the irradiation group (group I);3) high concentration group(group H);4)middle concentration group (group M) and5) low concentration group(group L). Afterirradiation for24,48and72h, cell proliferation was measured by MTT. The expressionof α-smooth muscle actin(α-SMA) in cell was observed by Immunocytochemical stainingand the expression of α-SMA, collagen type I and III were detected by western blot.Results1In the range of5~15Gy, X-ray could promote cell proliferation of HELF,and the best dose was5Gy which had a statistical significance with control group andother radiation groups(P<0.05).2Compared with control group in the same time point,cell proliferation activity was increased in the irradiation group in24,48and72h andhad a statistical significance between control and radiation group(P<0.05).3Theexpression of collagen type I was increased for2.54,2.69and3.75folds respectively in24,48and72h radiation group, compared with control group in the same time point.Like expression of collagen type I, the collagen type III also increased1.52,2.18and3.14folds. There had a statistical significance between control and radiationgroup(P<0.05).4The cell proliferation increased by X-ray could be inhibited bytreatment with Dong qin qing jin mixture.5The collagen type I and III were up-regulation for4.38and4.93folds respectively in irradiation group and prevented to20% and23%respectively in Qin qing jin mixture group(P<0.05).6. Treatment with Dong qinqing jin mixture could attenuated positive expression of α-SMA in MRC-5cells inducedby X-ray, and inhibited the increasing expression of α-SMA, collagen type I and III. Allthe differences were statistically significant (P<0.05).Conclusions1The radioactive embryonic lung fibroblasts proliferation model could bemade with irradiated in a5Gy dose for72h;2Dong Qin qing jin mixture could inhibitthe cell proliferation activity and the expression ofα-SMA, collagen type I and IIIinduced by X-ray irradiation. |
