Objective: To explore the protective effect of sericin on the type2diabetes by measuring changes of the blood glucose level, plasma insulinand glucose tolerance in the streptozocin(STZ) combined with high-fat dietinduced mouse model of type2diabetes. Mehods: The diet-inducedobesity(DIO) mice were interaperitoneally injected STZ(70mg/kg) toinduce type2diabetic models.40diabetic model mice(BG>199.8mg/kg)were randomly divided into5groups: control group, positive controlgroup(rosiglitazone:20mg/kg/d), and sericin treated groups(group: A, B,C), with8animals in each group. Animals in group A, B, C wereadministered different doses of sericin by PO(A:0.3g/kg/d, B:1.0g/kg/d, C:3.0g/kg/d). All of the animals were administered with ddH2O orcorresponding compounds for28days. The blood glucose(BG) wasmeasured once a week, while the body weight(BW) and food intake(FI)was measured twice a week. OGTT was performed on day14and day28.Plasma insulin and triglyceride(TG) level were measured with the ELISAand glycerol lipase oxidase(GPO-PAP) respevtively. At the end of study,the organ coefficients were measured, and islet pathological sections werestained through immunohistochemistry. Result:1, At the end of study,compared with the control group, the BG of group B, group C and positivecontrol group was significantly lower(P<0.05).2, The BW and FI of thetreatment groups have no significantly difference, except for the positivecontrol group.3, On day14, compared with the mice in control group, theAUC0~120minof mice in group B, group C and positive control group wassignificantly lower(P<0.05). But there had no significantly differencebetween group A and the control group on day28.4, Compared with control group, insulin level of mice in group C was significantly higher,while there was no significant difference in other groups(P<0.05).5,Compared with the mice in control group, the TG of mice in group C andpositive control group was significantly lower(P<0.05).6, There was nosignificant difference among the treatment groups and the control group inkidney coefficients and liver coefficients. The fat coefficients of mice inpositive control group was significantly higher(P<0.05) than that of micein control group.7, Compared with control group, the insulin positivereaction is more obvious of islet pathological sections in group C.Conclusions: Sericin can improve β-cell function and promote thesecretion of insulin to decrease BG and TG, to improve the glucosetolerance. It is concluded that sericin has protective effects on type2diabetic obese mice.
