ObjectiveCerebrovascular disease is a common disease in Middle-aged people and the old. Edaravone is a free radical scavenger, monosialoganglioside resistance to neural cell apoptosis. neural cell apoptosis is the main in ischemic penumbra after local cerebral ischemia/reperfusion. In recent years,PI3 K /Akt signal transduction pathway is high profile in adjusting the neuron apoptosis after cerebral ischemia/reperfusion.To investi-gate the effects of Monosialoganglioside and Edaravone on expressions of PI3K、Akt、PDK1 and GSK3βprotein in ischemic penumbra after local cerebral ischemia /reperfusion in intraluminal thread occlusion of the middle cerebral artery(MCAO) rats and its related mechanism. To provide experimental basis for clinical reasonable application. MethodsThe local cerebral ischemia/reperfusion model was established by intraluminal thread occlusion of the middle cerebral artery. The animals were randomly divided into pseudo surgery group(sham group), model(MCAO)group, Monosialoganglioside group, Edaravone group and Monosialoganglioside and Edaravone group. using the techniques of immuno-histochemical sraining,the expressions of PI3K、Akt、PDK1 and GSK3βprotein was observed at 3,7 and 14 days in ischemic penumbra. Image analysis system for Nanjing Jieda was used, statistical methods for the variance analysis and T test was applied. Results1)A value and positive unit of PI3K、Akt protein expression in model group were higher than those in sham group in ischemic penumbra after local cerebral ischemia/reperfusion in MCAO rats( P<0.01). A value and positive unit of the expressions in Monosialoganglioside and Edaravone group were higher than those in MCAO rats(P<0.01).For most of a value and positive unit of the expressions in Monosialoganglioside and Edaravone groups were higher than those in MCAO rats(P<0.01).2)A value and positive unit of PDK1、GSK3β protein expression in model group were higher than those in sham group in ischemic penumbra after local cerebral ischemia/reperfusion in MCAO rats( P<0.01). A value and positive unit of the PDK1 protein expression in Monosialoganglioside and Edaravone group were higher than those in MCAO rats(P<0.01);A value and positive unit of the GSK3β protein expression in Monosialoganglioside and Edaravone group were lower than those in MCAO rats(P<0.01). a value and positive unit of the PDK1 protein expression in Monosialoganglioside and Edaravone groups were higher than those in MCAO rats(P<0.01);For most of a value and positive unit of the GSK3β protein expression in Monosialoganglioside and Edaravone groups were lower than those in MCAO rats(P<0.01). Conclusions1) The expression of PI3K、Akt、PDK1and GSK3β protein were higher than those in ischemic penumbra in MCAO rats,PI3 K /Akt signal transduction pathway was activated.2)Monosialoganglioside and Edaravone resist neural cell apoptosis, regulate PI3 K /Akt signal transduction pathway by enhancing PI3K、Akt and PDK1 protein and restraining GSK3β expression after local cerebral ischemia /reperfusion in MACO rats.
