BackgroundAccording to the survey of world health organization (WHO), cerebrovascular disease is one of the three major diseases that resulted in the deaths of human, is also of the world’s fourth largest disease leading to lose labor ability. There are all kinds of biochemical markers can be used to detect pathological state, such as infections, diabetes, and dyslipidemia, but there was no specific blood biochemical markers can be used in the monitoring of cerebrovascular disease. Microparticle is the capsule bubbles off from various cell surfaces in the circulation system with diameter between 0.1 and 1 um. Peripheral circulation microparticles can be produced by various types of cells, such as platelets, red blood cells, endothelial cells and smooth muscle cells, and so on. Many studies showed that the membrane surface of microparticles carry specific antigen and CD molecules of its parent cell. Previous studies found that the amount and type of circular membrane microparticle is closely related to many diseases, such as high blood pressure, coronary heart disease, atherosclerosis and many other diseases. At the same time, microparticles are closely related to inflammatory response, apoptosis and other pathological physiological process. Endothelial microparticles originate from endothelial cells, also act as a symbol of reflect endothelial cell function.ObjectiveIn this experiment, we use the method of flow cytometry, to investigate circulating levels and antigen expression of microparticles in ischemic cerebrovascular disease; also to explore the correlation between ischemic cerebrovascular disease and microparticles level. To further study on the microparticle’s effects on vascular endothelial cell function, accordingly to further explore the role of microparticles in mechanism of ischemic cerebrovascular disease.Material and methodsIn this experiment, circular microparticles separation extracting from venous blood of patients with the first occurrence of ischemic stroke is acting as the research object. At the same time, circular microparticles extracting from venous blood of the healthy individuals for physical examination is acting as a controlled group. Two groups of microparticles were combined with fluorescent antibody, completed the antigen type tag after incubation of cells in dark, and quantitative detection of the amount and type of microparticles by flow cytometry.Results1.Circulating endothelial microparticles:in the experimental group, number of CD 144+ CD41a-, CD31 + CD41a-, CD62E+, and AnnexinV+ CD62E+ endothelial microparticles are higher than that of the control group, the difference between the two groups have statistical significance.2.The circulating platelet microparticles:there was no statistically significant difference in the number of circulating platelet microparticles in the experimental group and control group.3.The correlation of circulating microparticle number and ischemic cerebral infarction:the number of CD 144 + CD41a - endothelial microparticles was associated with the severity of the stroke, the correlations were positively correlated; and the levels of AnnexinV + CD62E + endothelial microparticles were in moderate correlation with subtypes of stroke; the rest of the types of endothelial microparticles showed no relevance to stroke.Conclusion1.There is no difference in the number of circulation platelet microparticles in ischemic cerebral infarction patients and healthy people;2.The circulating endothelial microparticle level is significantly increased in ischemic cerebral infarction patients relative with healthy people, which may indicated that endothelial microparticles may have impact on endothelial cell function, and promote the development of the occurrence of stroke;3.CD 144 + CD41a - endothelial microparticles may be as prompt effective biomarker of the severity of ischemic stroke.
