To Investigate The Diuresis Function In Rats By Study At The Expression Of AQP1&2

Objective:To observe the Jian Pi Yi Shen acupuncture effects of common conditions and diuresis functions in rats.1. To observe the Jian Pi Yi Shen acupuncture effects of urine electrolytes in rats.2. To observe the Jian Pi Yi Shen acupuncture effects of AQP1&2 expression in rats’kidney and urine; and disuse the relationship between kidney and urine of the AQP1&2 expressionMethod:3 months only 45 healthy male SD rats were randomly divided into control group, hydrochlorothiazide positive control group, the Tonifying Spleen and Kidney Acupuncture group.The blank control group and the Tonifying Spleen and Kidney Acupuncture group are received gavage in a dosage of 0.9% saline 10 ml/kg, the Tonifying Spleen and Kidney Acupuncture group acupuncture Shen Shu, Da Zhu, Zu Sanli. Hydrochlorothiazide positive control group are received gavage in a dosage of 30% hydrochlorothiazide 10 ml/kg. All these three groups were received three mouths intervention. Test weight every week. Collect urine of rats with urine (?) collected urine of rats by PVC pipes. Then sent the pipes of urine to clinical laboratory of Xin Xinhua hospital for testing the urine. When beheaded the rats, remove the rat kidney tissue into liquid nitrogen for preserved. Using western blot and Q-polymerase chain reaction (PCR) to detect the expression of AQP1、AQP2 and mRNA.Result:1. common conditions of all groups of rats, weight, diet and mind are no significant difference before the interruption. After the experiment, urine volume had statistical difference between acupuncture group and control group, and urine volume had high statistical difference between HCTH group and control group.2. Urine electricity had no statistical difference between acupuncture group and control group. The electricity K+ had statistical difference between HCTZ group and control group, and The electricity Na+ had statistical difference between HCTZ group and control group.3. Acupuncture and HCTZ can decrease the express of kidney AQP1 and mRNA. But express of kidney AQP1 had no statistical difference between acupuncture group and control group. Acupuncture and HCTZ can lower the express of kidney AQP2 and mRNA. The express of kidney AQP2 had no statistical difference between acupuncture group and control group, and had high statistical difference between HCTZ group and control group.4. Acupuncture and HCTZ can increase the express of urine AQP1 and mRNA. The nt ratio had statistical difference between acupuncture group and control group, and had high statistical difference between HCTZ group and control group. Acupuncture and HCTZ can increase the express of urine AQP2 and mRNA. The nt ratio had high statistical difference between acupuncture group and control group, and had high statistical difference between HCTZ group and control group.Conclusion:1. The research shows that, compare with HCTZ group, the common conditions of acupuncture group had no obvious effect. To do the tonifying spleen and kidney acupuncture with Shen Shu, Da Zhu, Zu Sanli acupionts, would increase the urine volume of water heavy rats.2. The diuresis effect of acupuncture would not influence the urine electrolyte. But HCTZ had natriuretic effect. It will cause electrolyte disturbances if take it in usual.3. The diuresis effect of acupuncture was correlative with the express of kidney AQP1&2. We infer that acupuncture could inhibit tubular reabsorption by increasing expression of kidney AQP1&2.4. Acupuncture could increase the expression of urine AQP1&2. The expression of kidney AQP1&2 was correlative with urine AQP1&2.
