The Expression Of GPâ…¡B/â…¢A In Rats With Early Fatty Liver Model And The Intervention Effect Of Gexia Zhuyu Decoction

Objective:By copying the early nonalcoholic fatty liver disease (NAFLD) model in rats, to detect the expression level of platelet membrane glycoprotein â…¡B/IIIA(GP II B/IIIA) dynamically for making it clear that whether there is a phenomenon of platelet activation in early NAFLD. Simultaneously, using Gexia Zhuyu Decoction which is one of the classical decoction in promoting blood circulation for removing blood stasis to intervene for making it clear that its effect on the expression of GP II B/IIIA in early NAFLD. Preliminary revealed the relationship between the early NAFLD and blood stasis to provide the theory evidence and the basis of experimental research for clinical effective treatment of TCM.Methods:We divided 180 male SD rats randomly into normal group I and model group I for each 50,and normal group II, model group II, simvastatin group and Gexia Zhuyu Decoction group for each 20. Besides the normal group I and normal group II, other groups were feed with high fat fodder to copy the model. Since the weekend of 8th, normal group II and model group II were given intragastric administration with saline, simvastatin group and Gexia Zhuyu Decoction group were given intragastric administration with drugs. At the weekend of 4th,6th,8th,10th,12th, respectively detected the body weight, liver wet weight, the ration between liver weight/body weight, liver enzyme (ALT, AST), blood lipid and liver lipid (TC, TG) and platelet aggregation rate and the expression level of GP â…¡B/IIIA (ELISA) of rats in different groups. Paraffin section (HE staining) and frozen section (Sudan â…¢ dyeing) to observe the fatty degeneration of liver cells.Results:1. At the weekend of 4th,6th,8th,10th,12th,the body weight, liver wet weight, and the expression level of GPâ…¡B/â…¢A in model group â…  are higher than those in normal group â… , the difference between them have statistical significance. The blood lipid (TC)and liver lipid (TC, TG) in model group â…  are higher than those in normal group 1, the difference between them have statistical significance. At the weekend of 12th, liver enzyme (ALT, AST) in model group â…  are higher than those in normal group â… , the difference between them have statistical significance. At the weekend of 6th,8th,10th,12th, the ration between liver weight/body weight and the blood lipid (TG) in model group â…  are higher than those in normal group â… , the difference between them have statistical significance. At the weekend of 8th,10th,12th,the platelet aggregation rate in model group â…  are higher than those in normal group â… , the difference between them have statistical significance.The observation on fatty degeneration of liver cells in model group â…  on pathological morphology indicates the model are copied successfully.2. Liver enzyme (ALT, AST):At the weekend of 12th,the difference between model group â…¡ and normal group â…¡, has statistical significance,Gexia Zhuyu Decoction group and model group â…¡ have statistical significance.Blood lipids (TC, TG):At the weekend of 10th and 12th, the difference between model group â…¡ and normal group â…¡ has statistical significance, the difference between simvastatin group,Gexia Zhuyu Decoction group and model group â…¡ have statistical significance;Liver lipid (TC, TG):At the weekend of 10th and 12th,the difference between model group â…¡ and normal group â…¡ has statistical significance.At the weekend of 12th,the difference between Gexia Zhuyu Decoction group and model group â…¡ has statistical significance.Platelet aggregation rate:At the weekend of 10th and 12th,the difference between model group â…¡ and normal group â…¡ has statistical significance.At the weekend of 10th,the difference between Gexia Zhuyu Decoction group and model group II has statistical significance. At the weekend of 12th,the difference between simvastatin group,Gexia Zhuyu Decoction group and model group II have statistical significance.Platelet membrane glycoprotein GP â…¡b/â…¢a:At the weekend of 10th and 12th, the difference between model group II and normal group II has statistical significance.The difference between simvastatin group,Gexia Zhuyu Decoction group and model group II have statistical significance.Morphological changes:At the weekend of 10th and 12th,liver tissues stained by HE staining and Sultan III shows that a large number of liver cells appeared fatty degeneration in model group â…¡, simvastatin group and Gexia Zhuyu Decoction group compared with model group II, the degree of fatty degeneration of liver cells are mitigated.Conclusion:1. The expression level of GP â…¡b/â…¢a are obviously increased in early NAFLD model rats.It shows the phenomenon of platelet activation is existed in the process of the early NAFLD.2. Gexia Zhuyu Decoction can improve the condition in liver enzyme,blood lipid and liver lipid (TC, TG) and the degree of fatty degeneration in liver cells. It also can inhibit the platelet aggregation rate and the expression level of GP â…¡B/â…¢A, and shows blood stasis is existed in the process of the early NAFLD.
