Purpose:1. To construct rat model with venous hypertension which can induce DAVF.2. Detection of Ephrin B2 and its expression in the dural venous hypertension rat model, the distribution of space-related angiogenic factors, explore the possible role of Ephrin B2 in dural arteriovenous fistula formation process.Methods:1.Firstly:The establishment of venous hypertension rat model; 60s SD rats were randomly divided into three groups, which group A deal with right carotid artery and external jugular vein anastomosis, In addition to ligate the superior sagittal sinus and outway of the left side of transverse sinus vein; group B ligate the superior sagittal sinus and outway of the left side of transverse sinus vein; group C were sham group; at the same time taking measurements of drain venous pressure (DVP). After 12 weeks, all rats recived abdominal aortic cannulation cerebral angiography to observe whether or not the formation of dural arteriovenous fistula2. Part II:120s SD rats were randomly divided into three groups, which group A deal with right carotid artery and jugular vein anastomosis, In addition to ligate the superior sagittal sinus and outway of the left side of transverse sinus vein; group B ligate the superior sagittal sinus and outway of the left side of transverse sinus vein; group C were sham group;10 rats were killed each of the groups respectively in 1,3,7,14 days after the surgery, of which four deal with immunohistochemistry, tissues detect the space distribution of Ephrin B2 in rat brain dural,6 rats were taken real-time PCR to detect the expression of Ephrin B2 mRNA.Results:1. Part â… :group A of the rats draining venous pressure with an average of 20.8±2.2mmHg, significantly higher in group B 5.5±0.9mmHg mmHg, group C was 1.9±0.3mmHg. Statistical analysis:P<0.01;LSD pairwise comparison method row, there were different between the groups.2. Part â…¡:Postoperative day 1, EphrinB2 highly expressed in endothelial cells where visible dural immunohistochemical staining in group A and B, and reach peak in 7 days after sugury, day 14 began to decline, IOD value was significantly higher at day 7 between group A and group B (P<0.05); group C, there was no significant EprinB2 expression. qPCR results, compared with the surgery group, at the 1st day after surgery, EprinB2 mRNA transcript levels increased in group A, and peaked in 7st days, days 14 began to decline, group B is similar to the A group; HIF-1α mRNA transcript in the at Part I in group A, and decreased with time; the transcription of EprinB2 and HIF-la showed some correlation.Conclusion:Venous hypertension model can induce stable DAVF, the induction rate is about 40%. ephrin B2 began to rise in postoperative days 3,7 days reach the peak, began to decline between 7-14 days.
