| Objective: To inspect the expression of mi R-143 and P53/K-ras in normal colon tissue,colorectal adenomas tissue and the colorectal cancer tissue for exploring the role of mi R-143 and P53/K-ras in development of colorectal cancer, and to provide a theoretical basis for pathogenesis and early discovery of patients with colorectal cancer.Methods : P53 and K-ras were detected by immunocytochemistry(IHC) and Western blot in normal colon tissue,colorectal adenomas tissue and the colorectal cancer tissue,and each of them were 9 cases.Mi R-143 was detected by q RT-PCR technology in the above same cases.Results:(1)The results of IHC detection showed that the positive expression rates of P53 protein is gradually increased in the normal colon, colon colorectal adenomas and the colorectal cancer groups, respectively 0%(0/9), 22.2%(2/9), 100%(9/9).There was no significant difference in the positive rates of P53 expression between normal colon and colorectal adenomas groups(P>0.05).Meanwhile,there was significant difference in the positive rates of P53 expression between colorectal cancer and normal colon groups, so were colorectal cancer and colorectal adenomas groups(P<0.05).The results of IHC detection showed that the positive expression rates of K-ras protein is gradually increased in the normal colon, colorectal adenomas and the colorectal cancer groups,respectively 11.1%(1/9),66.7%(6/9),100%(9/9).There was no significant difference in the positive rates of K-ras expression between colorectal adenomas and the colorectal cancer groups(P>0.05). Meanwhile, there was significant difference in the positive rates of K-ras expression between normal colon and colorectal adenomas groups, so were colorectal cancer and normal colon groups(P<0.05).(2)The results of Western-blot showed that expression of P53 protein is gradually increased in the normal colon, colorectal adenomas groups,colorectal cancer groups, respectively 0.223±0.053,0.298±0.010,0.785±0.166. There was significant difference in the expression of P53 among these 3 groups( F=62.195,P<0.05). Multiple compared each other in these 3 groups, P53 expression in colorectal adenomas groups(0.298±0.010) was higher than that in normal colon groups(0.223±0.053), there was no significant difference between the two groups(P>0.05).P53 expression in colorectal cancer groups(0.785±0.16) was higher than that in colorectal adenomas groups(0.298±0.010),there was significant difference between the two groups(P<0.05), P53 expression in colorectal cancer groups(0.785±0.16) was higher than that in normal colon groups(0.223±0.053), there was significant difference between the two groups(P<0.05). The results of Western-blot showed that expression of K-ras protein is gradually increased in the normal colon, colorectal adenomas and the colorectal cancer groups, respectively 0.311±0.092,0.591±0.188, 0.735±0.105. There was significant difference in the expression of K-ras among these 3 groups (F=22.732,P<0.05). Multiple compared each other in these 3 groups, K-ras expression in colorectal cancer groups(0.735±0.105) was higher than that in colorectal adenomas groups(0.591±0.188), there was significant difference between the two groups(P<0.05). K-ras expression in colorectal cancer groups(0.785±0.16) was higher than that in normal colon groups(0.311±0.092), there was significant difference between the two groups(P<0.05); K-ras expression in colorectal adenomas groups(0.591±0.188) was higher than that in normal colon groups(0.311±0.092),there was significant difference between the two groups(P<0.05).(3)The results of RT-PCR showes that expression of mi R-143 in the normal colon, colorectal adenomas and colorectal cancer groups were 0.935±0.342, 0.144±0.103, 0.222±0.134. There were significant difference in the expression of mi R-143 among these three groups(F=35.227,P<0.05),Multiple compared each other in these 3 groups, mi R-143 expression in colorectal adenomas groups(0.144±0.103) was lower than that in colorectal cancer groups(0.222±0.134), there was no significant difference between the two groups(P>0.05).mi R-143 expression in colorectal adenomas groups(0.144±0.103) was lower than that in normal colon groups(0.935±0.342),there was significant difference between the two groups(P<0.05). mi R-143 expression in colorectal cancer groups(0.222±0.134) was lower than that in normal colon groups(0.935±0.342),there was significant difference between the two groups(P<0.05).(4) The correlation of mi R-143 and P53 protein, K-ras protein expression was analyzed by Spearman test. The results showed that there was no significant correlation between mi R-143 and P53 protein(P>0.05), and the relationship of mi R-143 and K-ras protein was negative(PP<0.01), Spearman correlation coefficient(R) were:-0.371,-0.792.Conclusions:1.The overexpression of P53 might be involved in the development of colorectal cancer,but it was not obvious to colorectal adenomas.2.The overexpression of KRAS might be involved in the development of colorectal adenomas and colorectal cancer.3.The expression level of mi R-143 decreased might be involved in the development of colorectal adenomas and colorectal cancer. 4.There was no significant correlation between mi R-143 and P53,suggesting that P53 might not regulate or not directly regulate the expression of mi R-143;The relationship of mi R-143 and K-ras was negative,suggesting that mi R-143 is involved in the occurrence and development of colorectal adenoma, colorectal cancer via regulating K-ras gene. |
