Object:The present study was to investigate the association between Estrogen Receptor 1 (ESR1) Gene single nucleotide polymorphism(SNP) and genetic susceptibility to hepatocellular carcinoma(HCC) in liver cancer family groups of Zhuang population in Fusui county of Guangxi province.Methods:Cohort study and polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) were adopted to detect the genotypes frequency of 85 members of 21 HCC high incidence families and 39 members of 10 control familiesof zhuang population in Fu sui county, Guangxi province. Correlation between the ESR1 gene polymorphisms and HCC risk was evaluated by non-conditional logistic regressions.Results:1. (1) The estrogen receptor1 gene rs3798757 genetic frequency of genotypes AA, AG, GG among the HCC high incidence families and controls was 83.53vs74.36%,11.76 vs 17.95% and 4.71 vs 7.69%, respectively.(2) For members of the HCC high incidence families, the oddsrations of suffering from HCC among people carrying AG,GG genotypes were 0.298 (95%CI= 0.035-2.515, P=0.266) and 0.671 (95%CI= 0.070-6.391, P= 0.729). But in control families, the oddsrations of suffering from HCC for the AG, GG genotypes carriers were 0.218 (95%CI= 0.025-1.917, P=0.170) and 0.509 (95%CI=0.049-5.260, P=0.571), respectively2. (1) The estrogen receptor 1(ESR1) gene rs3798758 genetic frequency of genotypes GG, GT and TT were 85.72%,9.52% and 4.76% in HCC-clustering families and 76.92%,17.95% and 5.13% in normal families.(2) In HCC high incidence families, the oddsrations of HCC for people with GT, TT genotypes were 0.281 (95%CI=0.059-1.344,P=0.112) and 0.597 (95%CI= 0.062-5.719, P=0.655). But for members of control families, the oddsrations of suffering from HCC for GT,TT carriers were 0.476 (95%CI= 0.089-2.546, P= 0.386) and 0.833 (95%CI=0.070-9.858, P=0.833).Conclusion:1. Genotype distributions of the estrogen receptorl gene rs3798757 and rs3798758 in both group people conformed to the Hardy-We inberg equilibrium.2. Neither ESR1 gene rs3798757 nor rs37987578 single nucleotide polymorphism has significant correlation with hepatocellular carcinoma for people in Fusui county of Guangxi province.
