| Coreopsis tinctoria Nuff., called "snow chrysanthemum" or "Kunlun chrysanthemum", which had a good health care function in terms of the metabolic syndrome, had attracted wide attention of scholars in recent 15 years. In order to better develop and take advantage of this excellent plant resource, the subject of chemical and pharmacological research was taken deeply and the quality of plant samples from different sources were explored.1. In this study, chemical research was carried out from the eluation of macr-oporous resin (D101).22 compounds were isolated and identified by traditional c-hemical techniques, modern chromatographic techniques and spectrum technology, including 3 chalcones,6 dihydroflavones,1 dihydroflavonol,2 flavonols,1 flavone, 2 aurones,1 benzenepropanoic acid ester,2 monoterpenoids,3 sesquiterpenoids, an-d 1 phenylethanoid glycoside. They were butein(1), okanin(2), marein(3),2S-7,3’, 5’-trihydroxyflavanone(4), butin-7-O-β-D-glucopyranoside(5), isookanin(6), flavanoma-rein(7),5,7,3’,5’-tetrahydroxyflavanone-7-O-β-D-glucopyranoside(8), pruning(9), taxif-olin(10), quercetin(11), isoquercitrin(12), luteolin(13), sulfuretin(14),7,3’,4’-tetrahyd-roxyaurone(15), linocinnamarin(16), (1S,2R,4R)-p-menth-8-ene-1,2-diol(17), trans-p-menthane-1,2,8-troils(18), coretinphenol(19), coretinterpenoid A(20),3β-glucopyranos-yloxy-β-ionone(21), phenylethyl-8-O-β-D-glucopyranoside(22). Compound 20 was a new bisabolane-type sesquiterpenoid and compounds 5,9,14,17,18,21 were iso-lated from this plant for the first time.2. In this study, glucose-induced insulin resistance HepG2 cell model was established to evaluate glucose absorption by pre-administrating compounds 3,7,8,16. The results showed that compound 3 at 6.25 μmol·L-1,12.5 μmol·L-1,25 μmol·L-1,50 μmol·L-1, compound 7 at 6.25 μmol·L-1,12.5 μmol·L-1,50 μmol·L-1, and compound 16 at 12.5 μmol·L-1,25 μmol·L-1 all exhibited good hypoglycemic activity in vitro. It proved that pre-administrating those compounds could be good prevention of insulin resistance.3. In this study, oleic acid and cholesterol,25-hydroxy cholesterol-induced fat accumulation HepG2 cell models were established to evaluate the effects of compounds 3,6,7,8,11,16,19 on TG and TC. The results showed that TG were significantly cleared by administrating compound 6ã€8ã€19 at 10 μmol·L-1 and TC were significantly cleared by administrating compound 7 at 10 μmol·L-1.4. In this study, compounds 2,3,6,8,10,16 were determinated on samples which from 6 different seed sources(Xinjiang, Jiangsu, Ningxia, USA) of Coreopsis tinctoria and 3 growing regions (Minfeng County, Xinjiang Province; Yanqing County, Beijing; IMPLAD, Beijing). The contents of these compounds were:3.69~7.91 mg·g-1, 21.72-72.68 mg·g-1,1.79~4.87 mg·g-1,1.44~11.27 mg·g-1,0.83~2.92 mg·g-1,0.18~0.46 mg·g-1 respectively. The contents of 3,8,16 were much more affected by seed sources and the contents of 2,6,10 were much more affected by growing regions.This research enriched the structures of compounds found in Coreopsis tinctoria and further revealed the material basis with good hypoglycemic and hypolipidemic activities. It also provided a reference of quality control on Coreopsis tinctoria. In conclusion, the research laid the foundation for further development and utilization of the plant resources. |
