Objective: To study the chemical constituents and water-soluble polysaccharide of Actinidia Chinensis Planch.and the primary biological activities. To make Actinidia Chinensis Planch. polysaccharide into granules, and to establish a method of quality control.Methods: The chemical constituents of Actinidia Chinensis Planch. were studied by preliminary test, and on the base of it, the silica gel, Sephadex LH-20, ODS, high preparative performance liquid chromatography were used to isolate and purify the chemical constituents. Their structures were identified on the basis of the spectra data and physicochemical properties. The content of polysaccharide was determined by the method of phenol-sulfuric acid, DPPH method and caenorhabditis elegans nematode were used to detect the antioxidative and anti-aging effects, high performance gel permeation chromatography(HPGPC) was used to determine molecular weight of the purified polysaccharide. On the base of these, single-factor method and the extraction efficiency were used to optimize the extraction process. Appearance, moisture absorption rate and molding rate were considered to screen out excipient and formulation process.Results: The results indicated that Actinidia Chinensis Planch. might contains amino acid, polypeptide, protein, saccharides, glycosides, saponins, organic acids, flavonoids, phenols and tannins, sterides, triterpenes. Eight components were identified as follows: 2α,3β-dihydroxy-urs-12-en-28-oic acid(AC-1), 2α,3α-dihydroxy-olean-12-en-28-oic acid(AC-2), 2α,3α-dihydroxy-urs-12-en-28-oic acid(AC-3), 3β-acetyloxy-ursolic acid(AC-4), ursolic acid(AC-6), olean acid(AC-7), β-sitosterol(AC-9), daucosterol(AC-10). After partial purification, the contents of polysaccharide were 74.54%. The results showed that Actinidia Chinensis Planch..Polysaccharide was actived on antioxidative and anti-aging effects. Purified polysaccharides were isolated which were named as ACP-2 and ACP-3, and molecular weight of them were 1079 k Da, 1031 k Da. The granule has the better molding rate when it with mannitol and 90% alcohol. Quality studies on the granule showed that grain size, moisture and dissolubility meet the stipulations. The average level of polysaccharide was 30.04%.Conclusion: Compounds AC-1、AC-2、AC-3、AC-4 were isolated from the fruits of Actinidia Chinensis Planch. for the first time. Actinidia Chinensis Planch. polysaccharide could be used as effective fraction. And it was active for oxidation resistance and anti- aging. The quality control of the capsule is reliable.
