| Objective To observe the change of tissues and shapes on hippocampus areas in myocardial ischemia rats through electric acupuncture with the yuan point and different matching acupoints, effect on the expression of brain-derived neurotrophic factor(BDNF) and tropomyosin receptor kinase B(Trk B); to discuss the central control mechanisms of the protective effects of brain damage in myocardial ischemia rats by acupuncture; to compare with the specificity of the different acupoints compatibility effect.Methods 80 healthy male SD rats, weight about 220±20g,were randomly selected 15 to sham-operated group, others rats were narcotized through adopting the left anterior descending coronary artery ligation method and copying the AMI(Acute Myocardial Ischemia) model. The rats were randomly divided into 4 groups: model group, “shen menâ€acupoint group, “shen menã€zhi zheng†acupoint group and “shen menã€xin shu†acupoint group, each group have 15 rats. Conventional connection Powerlab 16 physiological recorder, electrocardiogram(ECG) of standard II lead records, Operation in the process of monitoring the heart rate and ECG ST segment and T Wave change. The model preparation in two days later, three electric acupuncture groups will start treatment. Stimulus current to 1 m A, frequency of 2 Hz, stimulate 15 min each time, once per day, for a week. Using HE staining to observe each rat hippocampus morphology, using Immunohistochemical method and real-time fluorescence quantitative PCR method to detect 5 groups of rat hippocampus in the expression of BDNF and Trk B. Data analysis by SPSS 17.0 for Windows.Results 1.Powerlab electrocardiogram: Compared with the sham-operated group, themodel rats’ heart rate was significantly declined, the ST segment was significantly raised, and the T wave was invasion. After the first day’s acupuncture, the rats’ heart rate of the “shen menâ€acupoint group, “shen menã€zhi zheng†acupoint group and “shen men ã€xin shu†acupoint group were higher than the model group, the difference was statistically significant(P < 0.01), prompt the electro acupuncture could improve the heart rate of myocardial ischemia rats. In the aspect of improving the heart rate, “shen menã€zhi zheng†acupoint group and “shen menã€xin shu†acupoint group were superior to the “shen menâ€acupoint group, the difference was statistically significant(P < 0.05). Between the “shen menã€zhi zheng†acupoint group and “shen menã€xin shu†acupoint group, there was no significant difference(P > 0.05). 2. Hippocampal neurons of each group: The rats’ hippocampal neurons, which in the sham-operated group were stained uniformity under the optical microscope, the neurons arranged well and integral; the rats of the model group, the neurons were arranged sparsely and loosely; the hippocampal neurons pathological changes were significantly weakened in the “shen menâ€acupoint group; the “shen menã€zhi zheng†acupoint group and “shen menã€xin shu†acupoint group’ s hippocampal neurons structure and arrangement compared with the model group had obvious improvement, cellular structure tend to be normal. 3. The BDNF and Trk B Immunohistochemical expression of rats hippocampus: Both groups of BDNF and Trk B had observed positive cells expression, most of them were the form of cytoplasm expression. To compare with the sham-operated group, the rats’ hippocampus of the model group of BDNF and Trk B immune response to positive neurons and the average optical density value decreased significantly, statistically significant difference(P < 0.05); to compare with the model group, the hippocampus of BDNF and Trk B in three group immune response to positive neurons and the average optical density value increased significantly, statistically significant difference(P < 0.01). 4. The BDNF m RNA and Trk B m RNA expression of rats hippocampus: Comparedwith the sham-operated group, the BDNF m RNA expression of rats hippocampus were increased in the model group, but no statistical difference(P>0.05).Compared with the model group, acupuncture can improve the myocardial ischemia rat hippocampus BDNF m RNA expression(P<0.05), “shen menã€zhi zheng†acupoint group and “shen men 〠xin shu†acupoint group were superior to the “shen menâ€acupoint group(P<0.01), “shen menã€xin shu†acupoint group are superior to “shen menã€zhi zheng†acupoint group,but no statistical difference(P>0.05), prompt acupoints compatibility is superior to the single acupoint when improved myocardial ischemia rat hippocampus BDNF m RNA expression levels;Compared with the sham-operated group, the rats hippocampus of Trk B m RNA expression were increased in the model group, but no statistical difference(P>0.05).Compared with the model group, acupuncture can improve the myocardial ischemia rat hippocampus Trk B m RNA expression(P<0.05), “shen menã€zhi zheng†acupoint group and “shen menã€xin shu†acupoint group were superior to the “shen menâ€acupoint group(P<0.01,P<0.05), “shen menã€xin shu†acupoint group are superior to “shen menã€zhi zheng†acupoint group,but no statistical difference(P>0.05).Conclusion 1. Acupuncture can improve cardiac function in myocardial ischemia of rats,meanwhile,acupuncture treatment can promote the recovery of cardiac function.Acupuncture treatment between yuan-shu points and yuan-source points has synergistic effect. 2. Myocardial ischemia could give rise to changes in the form of the hippocampus in rats, though the therapy of the electro acupuncture, and promote the restoration of nerve cells; the electro acupuncture can increase the expression of BDNF and Trk B in myocardial ischemia rat hippocampus, this may be one of the mechanisms of acupuncture against myocardial ischemia brain damage. 3. Acupuncture on myocardial ischemia rat damage protection has relative specificity, accommodation of the “shen menã€xin shu†acupoint group was superior to the “shenmenã€zhi zheng†acupoint group. Compatibility and synergy effect of acupoints had the relative specificity, yuan-shu points,yuan-source points and collateral-points combination are superior to the therapy of single acupoint. This kind of the acupuncture therapy will better protect to the heart, and against myocardial ischemia brain damage. |
