| Cuttlebone,also known as cuttlefish bone, is a traditional Chinese medicinal material, which has several kind of biological activitys,such as protection of gastric mucosa injury, blood coagulation and hemostatic efficacy.But during processing of cuttlebone, cuttlebone is usually discarded, which resulted in a waste of resources. If new drugs or healthy food was developed according to cuttlebone’s biological activity, not only make which turn waste into treasure, but also produce better economic returns. In this experiment, cuttlebone is the main raw material. Use advanced analysis methods to detect the structure of cuttlebone, verify the protective effect of it on gastric mucosal injury, blood coagulation activity and make the tablets which can treat for stomach ulcers. The main contents are as follows:1ã€Study on structure and main content of cuttlebone.By useing environmental scanning electron microscopy, infrared spectroscopy, X-ray diffraction, inductively coupled plasma this paper detected and analyzed the structure of cuttlebone in order to initially explore the relationship between its structure and some biological activity. By useing SEM scanned the ventral side’porous layer of massive cuttlebone,and found that the whole structure looked like a house rack which was formed by alcove-like grid structure with lots of porous. But after crushing, its grid structure was completely destroyed, which indicated that the grid structure had no relation with its blood coagulation and resisting gastric ulcer activity.The infrared spectroscopy and X-ray diffraction’s analysis showed cuttlebone contained aragonite. Compared with the complete cuttlebone, its ventral side’porous layer had basically the same chemical composition, but contained different content of aragonite. Furthermore, by using inductively coupled plasma mass spectrometry, this paper detected 70 kinds of inorganic elements from cuttlebone which included large amounts of Ca, a small amount of Na, Sr, Fe, P, K, Mg, and trace amounts of Si, Zn, Cu, Al, Br, Ba, B, Mn, Ni and so on.2ã€Study on the protective effect of cuttlebone on gastric mucosal injury which was induced by ethanol.First, this paper built acute gastric mucosal injury model which was induced by ethanol in Wistar rats. And then Wistar rats were fed with different doses of cuttlebone by intragastric administration. Finally, by testing gastric mucosal injury’s area, gastric mucosal injury’s index, inhibition ratio of injury, the volume of gastric juice, pH, MDA, GSH, PEG2, MPO, T-SOD,comparing the protective effect of each group on ethanol-induced acute gastric mucosal injury. The results suggestted cuttlebone had a role in protecting the ethanol-induced gastric mucosal injury. The possible mechanism of action was that there were neutralization between calcium of cuttlebone and gastric acid which increased gastric juice’s pH and mitigated its erosion effects on gastric mucosal.At the same time,it could also adjust the content of NO in gastric mucosal tissue to strengthen gastric mucosa protective barrier, promote the synthesis of PEG2 of gastric juice, gastric tissue and serum in oder to strengthen the resistance ability of the mucosal tissue and promote the healing of ulcer, enhance the vitality of SOD, remove of oxygen free radicals and reduce lipid peroxidation, reduce the content of MDA in gastric tissue and serum to reduce the damage for gastric mucosa cells, reduce MPO’s activity in gastric tissue of varying degrees and participate in adjusting the inflammation of gastric mucosa injury,which may be another mechanism of the preventive effect on gastric mucosa damage.3^ Study on clotting effect of cuttlebone. By analyzing the effects of cuttlebone suspension on bleeding and coagulation time in different experimental mice to verify its effect of coagulation, exploreing the possible clotting mechanism of it and providing theoretical basis for the development of new cuttlebone drugs. The results showed that under the 250 g/L dose, cuttlebone group’s bleeding time and clotting time were shorter 11.45 min and 1.38 min than the saline control group, respectively. Each dose group of cuttlebone could significantly shorten the bleeding time and clotting time of mice and compared with normal saline control group, bleeding and clotting time of cuttlebone solid group to different degrees of decalcification were significant difference, which showed that they also had clotting activity and the cuttlebone without decalcified had the shortest clotting time and best coagulation effect. Therefore, cuttlebone not only has clotting activity but also had a dose-dependent manner. Although calcium carbonate’s clotting time and bleeding time were different with normal saline control group, and the difference was not significant. So it had no blood coagulation activity. Cuttlebone can significantly shorten the clotting time, suggestted cuttlebone clotting activity might be associated with the content and functions of various blood coagulation factors; Cuttlebone could significantly shorten the bleeding time, which implies cuttlebone might be related to the platelet count and function, blood capillary function, the number of tissue factor, tissue contraction force and so on.4ã€Study on cuttlefish bone’s hemostatic effect in vitro. Through the experiment in mice, hemostatic properties of cuttlebone was compared with other common hemostatic materials. Guinea pig model was established to evaluate cuttlefish bone powder on healthy skin or damaged skin’s irritation reaction of guinea pig back to explore its sensitization. Through the femoral artery hemostasis test of mouse, the hemostatic time of ultrafine cuttlebone was (1.21±0.11) min, which was faster than the Yunnan Baiyao and starch. Hemostatic effect of ultrafine cuttlebone was ideal. Through the single exposure experiment of back healthy skin and back damaged skin of guinea pig, the characteristics of non-irritating, no skin sensitization, high security of the ultrafine cuttlebone were found.5ã€Study on preparation process of cuttlebone tablet.By single factor experiment and orthogonal test,it optimized the factor conditions which could affect the quality of the tablet were optimized. The optimal preparation process of cuttlebone tablet.as follows: Maltodextrin 1.0 g, supplemented CCNa concentration of 3%, the addition amount of magnesium stearate of 0.2%. According to the optimal formula, we got the tablet with bright and clean appearance. In addition, The tablet had little fluctuate of tablet weight and short breakdown time. |
