| Objective: The experiment is aimed to searching for the association betwween the Vascular endothelial growth factor gene polymorphism and2 type Diabetic retinopathy.Methods: The study takes the form of case contrlo, 1.according to the standards of diabetic diagnosis which put forward by ADA in 2014,we randomly select 85 patients with 2 type Diabetic retinopathy and 90 patients with 2 type Diabetes mellitus in Ophthalmology and Endocrine Department, and 88 normal person are selected in the physical examination centre. All participant were divided into DR group,DM group and control group.We diagnose Diabetic retinopathy according to the result of Mydriasis funduscopy and Fundus fluorescence angiography,general informations,such as Gender,BMI,Age,Blood pressure,Course of disease and FBG was recorded.2. 3 ml blood was took from all participants in the morning,we use the Blood Gen Mini Kit to extract DNA from the blood.3. We detect the gene polymorphism of the Vascular endothelial growth factor through DNA sequencing.4. All data was tested the normality. We use the SPSS to analysis all the data.Measurement data was compared using t or t’-test,P<0.05 was considered significant. Chi-square analysis was used to test whether VEGF genotype distribution followed the Hardy-Weinberg equilibrium.Odds ratio was counted to evaluate the risk factors of 2 type Diaetic retinopathy.Results:1.The allelotype frequencies and the genotype frequencies of VEGF in the 3 group were flowed the Hardy-Weinberg equilibrium.2.The C/C, C/A,A/A genotypic frequencies in control group, DM group,DR group are 59.09%ã€34.09%ã€6.81%;61.11%ã€34.44%ã€4.44%;48.23%ã€36.47%ã€15.29%,The frequency of AA gene were significantly increased in DR group than DM group and control group,the statistical difference is considered significant(P<0.05).3.The morbidity of DM patients with VEGF rs699947 AA is 3.88 times of those patients with VEGF rs699947 CC+CA.Conclusion:there is a clear link between the VEGF rs699947 gene polymorphism and 2 type Diabetic retinopathy,the AA genotype may be a predisposing genes of DR. |
