Halophyte is a kind of natural wild plants which can live in saline-alkali and to produce. China’s salt plant is rich in resources, but the development and utilization of relatively weak. Then, clear coastal salt biological nutrient content and its variation in different growth period, in favor of Halophytes in the further development and utilization. This study compared the four kinds of salt representative plants which in Hebei Binhai area has a wide distribution and more content(Limonium,Nitraria tangutorum Bobr, Herba Suaedae Glaucae Suaeda glauc, Saline Seepweed), the main components of flavonoids were analyzed to fully develop the natural resources in halophytes provides a scientific basis.Using the ultrasonic method to extract the flavonoids of the leaves and other parts of four halophyte,took rutin as the standard and using ultraviolet spectrophotometry to determination of total content of flavonoids in the halophytes. Through the study,we found that all of the plants of Halophyte contains rich flavonoids, In addition to the Nitraria, the other three kinds of halophytes have reached the maximum flavonoid content in Florescence.The content of total flavonoids:Nitraria tangutorum Bob>Herba Suaedae Glaucae Suaeda glauc>Saline Seepweed>Limonium spp.The major flavonoids in four kinds of halohpytes were analyzed by high performance liquid chromatography coupled with electrospray ionization mass spectrometry(HPLC-ESIMS). The experimental results indicated that the flavonoids extracted from halohpytes and in the same conditions,flavonoids from the extraction were identified.Thirteen flavonoid glucosides were presumed respectively for Gallic acid、epicatechin、myricetin、rutin, luteolin、quercetin、isorhamnetin、kaempferol、genistein, epigallocatechin gallate, hyperin, baicalin, eriodictyol.and six unknown flavonoids. Establish a sensitive and specific method of HPLC for simultaneous determination of flavone glycosides which was identified. HYPERSIR C18 BDS(5μm × 250 mm × 4.6 mm); mobile phase was the mobile phase A methanol, mobile phase was the mobile phase B 0.1% formic acid water; flow rate was 0.8 m L /min; the detection wavelength was: 270 nm, 370 nm; column temperature was 30 ℃; injection volume was 10μL. Use gradient elution method.The test results showed that Rutin and Epigallocatechin gallate of Nitraria reached the highest content at germination, the highest gallic acid,luteolin,eriodictyol and Baicalin content sprout period. Other flavonoids content in the Florescence peak.In addition to epicatechin gallic acid in the Limonium sprout period is at its maximum, Gallic acid, Myricetin, rutin and Genistein in germination period reached maximum, the rest of the content of flavonoids in flowering reached maximum.quercetin and Kaempferol in Saline Seepweed and Herba Suaedae reached the highest content at Leaf expansion,the highest rutin, hyperoside, baicalin and epicatechin and epicatechin gallic acid content Florescence,gallic acid and Isorhamnetin in Herba Suaedae reached the highest content at sprout period and Saline Seepweed do not contain isorhamnetin.Using DPPH, hydroxyl radicals, ABTS, reducing power and lipid peroxidation ability of five kinds of methods to evaluate the antioxidant activity of total flavonoids extracted from four plants of halophytes.The main results were as follows:Total flavonoids extracts from Nitraria tangutorum Bob、Herba Suaedae Glaucae Suaeda glauc、Saline Seepweed and Limonium spp reducing power, free radical scavenging activity and the lipid peroxidation inhibition has a certain relationship with concentration that the higher the concentration, the stronger the reducing power, the better activity of scavenging free radicals,the higher ability of inhibiting lipid peroxidation.Four kinds of halophytes are all have strong scavenging effect to free radicals; The flowers of Limonium spp、Herba Suaedae Glaucae Suaeda glauc and the fruit of Nitraria tangutorum Bob have a good reduction ability; The flowers of Limonium spp showed excellent inhibition of lipid peroxidation.The antibacterial activity was evaluated by Oxford cup and double dilution.The total flavonoids of four Halophyte in different growth period had a certain of bacteriostatic effect to Escherichia coli and staphylococcus aureus. Except for Nitraria,the inhibition of total flavonoids extracted from Limonium,Suaeda and Seepweed on Staphylococcus aureus than inhibition on Escherichia coli.Total flavonoids of four kinds of halophytes experiment in mice spleen cell proliferation, spleen cell protection experiment and test the immune experiment, SOD and MDA to explore different immune activity of total flavonoids in halophytes.The results showed that the four kinds of halophytes total flavonoids have significantly promoted(P < 0.01) on lymphocyte in 5-100 ug/ml, and are associated with the increase of the concentration showed a trend of first increases and then.In terms of the highest increment rate:Saline Seepweed>Herba Suaedae Glaucae Suaeda glauc>Nitraria tangutorum Bob>Limonium spp;The Limonium spp and Saline Seepweed overall on the protection of cells is more prominent;Therefore, adding different concentrations of Halophyte plant flavonoids can reduce cell damage, restore the activity of SOD, decrease the content of MDA.Studing the four halophytes total flavonoids stability.The results showed that the p H, temperature,metal ion, oxidant and reducing agent have the different impact on Limonium and the flower of Limonium 、 Saline Seepweed 、 Suaedae 、 Nitrariaand the fruit of Nitraria for the stability. Which significantly affect the stability of alkaline, Cu2+, Fe3+ H2O2, oxidant and reductant Na2SO3 of four halophytes flavonoids.
