BackgroundsBreast cancer is one of the most common malignant tumor in women. The morbidity rank the first in North America, northern Europe and other countries. Although the incidence of breast cancer is not high in our country, the incidence trend is rising year by year in recent years,and the growth rate is the fastest.The aetiological agent of breast cancer is not clear yet,it is a generalized diseases of high heterogeneity.So the treatment is a kind of systemic treatment,which needs combination of local therapy and comprehensive therapy.In recent years, with the development of immunohistochemical technology, the classification method of breast cancer gradually become a way which based on the combination of molecular pathological characteristics of breast cancer tissue and TNM staging classifiction.Now the main clinical classification refer to the st. Gallen consensus,using immunohistochemical technique,based on the result of ER,PR,HER-2 and Ki-67,to classify the breast cancer into four types,luminal A type, luminal B type,HER- 2 expression type and the triple negative breat cancer(TNBC).The triple negative breast cancer is a type which the expression of ER,PR and HER-2 are negative.Compared with other types, endocrine therapy and targeted therapy treatment are useless.So the way of treatment is limited,which leads to poor prognosis.In additon,the TNBC has unique biological behaviour,such as younger disease occurrence,racial difference,fast progress,strong invasiveness, trend to brain metastasis and visceral metastasis.So the prognosis is very poor.We urge to start from molecular pathology to find out its special molecular immunology marker to develop the targeted therapy drugs.At present, the biological behaviour of EGFR and E-cad protein in tumor occurrence, development, invasion and metastasis is studied more and more frequently.And relevant documents, in three negative breast cancer, EGFR into a high state, on the contrary, E-cad into a state of lower expression..This study aims to study the expression of two proteins in TNBC.By the comparison of biological characteristics to speculate their function in TNBC during tumor invasion and metastasis to provide a new way and basis to the targeted treatment of TNBC. ObjectivesTo explore the expression of EGFR and E- cad protein in triple negative and non triple negative breast cancer tissue to find out if there is difference,combined with their biological characteristics.To study their function in triple breast cancer during the occuring, progress, invasion and metastasis of tumor, to speculate whether they could be judgment factors for prognosis of TNBC. MethodsThis study collected all the tissue samples of patients having breast cancer which have complete clinical data and have been treated in the second department of breast surgery of the First Affiliated Hospital of Zhengzhou University between December 2012 and December 2014(wax block on file of pathology department being included). According to the results of immunohistochemistry,the patients was divided into two groups,the triple negative group and the non-triple negative group.Randomly selected 35 cases from triple negative groups as the experimental group, at the same time according to the proportion of 1:1,selected 35 cases in the non-triple negative group as control group.We used immunohistochemical S- P method to explore the expression of EGFR and E- cad protein in tumor tissues. The data was analyzed by SPSS 17.0 statistical software and statistical significance was tested by chi-square test,in which α=0.05. Results1.The positive expression rate of EGFR in triple negative group was obviously higher than that in the non-triple negative group, respectively was 37.1%(13/35) and 14.3%(5/35), the difference had statistical significance,(P < 0.05).E- cad in the triple negative group was significantly lower than that in the non-triple negative group, respectively was 25.7%(9/35), 51.4%(18/35), the difference had statistical significance(P < 0.05). 2. In 35 cases of triple negative breast cancer, EGFR expression was not related to age and menopausal status, had no significant correlation with tumor size and clinical stage(P > 0.05),but had significant correlation with histological grade of breast cancer and the status of lymph nodes metastasis(P < 0.05). 3.In 35 cases of triple negative breast cancer,the negative expression of E-cad was not significantly related to age and menopausal status(P > 0.05).But it was significantly related with tumor size, clinical stages,histological grade and lymph node metastasis status(P < 0.05). Conclusions1. E- cad and EGFR in the group triple and non-triple negative breast cancer were differentially expressed, and the difference was statistically significant.2.That EGFR expression rate was higher in triple negative breast cancer tissue, and E- cad expression rate was lower, was significantly related to TNBC’s clinical pathological characteristics,such as histological grade and axillary lymph node metastasis status.To determinetriple negative breast cancer has reference value.
